Objective:1.MRI Pfirrmann grading assessment and histological degeneration score(HDS)were used to evaluate the degenerative disc grading,and the consistency of the two methods was analyzed.The intrinsic relationship between MRI findings and histopathological changes in intervertebral disc degeneration was preliminarily investigated.contact.2.The role of mechanically sensitive ion channel protein Piezo1 in intervertebral discs with different degrees of degeneration was observed and examined,and its role in human intervertebral disc degeneration was preliminarily investigated.Methods: The intervertebral disc tissue specimens of 35 patients who underwent surgical resection of lumbar vertebrae from January 2017 to June 2018 were collected as experimental subjects.They were divided into two parts:1.The Pfirrmann grading system was used to evaluate the degree of degeneration of the lumbar sagittal image of the patient's preoperative MRI T2-weighted image before operation.HE staining of the intervertebral disc tissue obtained during operation was performed,and a modified version of histology was used.The degenerative score(HDS)standard was used for histopathological evaluation and consistency analysis was performed for both assessment methods.2.According to the evaluation results of intervertebral disc degeneration,the location and expression of Piezol protein in nucleus pulposus cells were detectedby immunohistochemistry(IHC),RT-PCR and Western-blot assay.The method was used to evaluate the apoptosis in nucleus pulposus cells.On the basis of this,the correlation between the expression of Piezo1 protein and apoptosis of nucleus pulposus cells in different degenerative groups was further analyzed.Results:1.There were no significant differences in preoperative general data between 1.35 patients(P>0.05),and Pfirrmann grade scores were consistent with HDS(P<0.05).2.There are Piezo1 protein expression in the nucleus pulposus cells of different degrees of degeneration.As the degree of degeneration increases,the expression level of Piezo1 protein increases,and there is a positive phase with the Pfirrmann classification(r=0.798).There was a positive correlation between the expression of Piezo1 protein and the apoptosis rate of nucleus pulposus cells(r=0.708).Conclusion:1.Pfirrmann grading is an excellent evaluation method for disc degeneration,whIHC can reflect the pathological changes of intervertebral disc degeneration to some extent.2.The expression of Piezo1 protein in nucleus pulposus cells is related to the degree of degeneration of intervertebral disc.Piezo1 protein may promote the process of intervertebral disc degeneration by promoting apoptosis of nucleus pulposus cells.High expression of Piezo1 protein may play a key role in disc degeneration associated with abnormal mechanical stress.
