Objective:In this study,the correlation among NLRP3,MDA,SOD and FEV1%pred was analyzed by detecting the levels of SOD and MDA in the serum of COPD patients as well as the expression of NLRP3 mRNA in peripheral blood lymphocytes,so as to explore the effect of oxidative stress mediated activation of NLRP3 inflammatory bodies on COPD.Methods:Selection of changsha first hospital from June 2017 to June 2018,90 cases of COPD patients in hospital,including 60 patients with AECOPD,according to pulmonary function in patients with AECOPD group is divided into weight-very restructuring and light-medium group,30 cases in each group,and reached the level remission after treatment symptoms and signs of stabilization of 30 patients with COPD and other selected physical examination as a control group of 30 cases of healthy subjects.Peripheral venous blood was collected from each group of subjects on an empty stomach in the morning.Serum levels of SOD and MDA were detected by ELISA.The expression of NLRP3 mRNA in each group was determined by RT-PCR.Results:1.MDA level and NLRP3 mRNA expression in serum of patients in AECOPD group and COPD stable group were higher than those in healthy control group(P<0.05).The serum SOD levels in AECOPD group and COPD stable group were lower than those in healthy control group(P<0.05).The serum MDA level and NLRP3 mRNA expression level of peripheral blood lymphocytes in AECOPD group were higher than those in COPD group(P<0.05).The serum SOD level in AECOPD group was lower than that in COPD stable group(P<0.05).2.In the AECOPD group,MDA level and NLRP3 mRNA expression level in the recombinant serum of severe to severe pulmonary function and peripheral blood lymphocytes were higher than those in the mild to moderate group(P<0.05).In the AECOPD group,the SOD level in the recombinant serum was lower than that in the light-medium group(P<0.05).3.Pearson correlation analysis:The serum MDA level in COPD patients was positively correlated with NLRP3 mRNA expression,and the correlation coefficient r was 0.64(P<0.05).There was a negative correlation between serum SOD level and NLRP3 mRNA expression in COPD patients,and the correlation coefficient r was-0.90(P<0.05).In COPD patients,FEV1%pred was negatively correlated with NLRP3 mRNA expression,and the correlation coefficient r was-0.78(P<0.05).Conclusion:1.The serum MDA level and NLRP3 mRNA expression level of peripheral blood lymphocytes were increased and SOD level was decreased in AECOPD group and COPD stable group.Oxidative stress and NLRP3 inflammatory corpuscule are involved in the pathogenesis of COPD.2.In the AECOPD soup,MDA level,NLRP3 mRNA expression in peripheral blood lymphocytes and SOD level were increased and decreased in the patients with severe to severe recombinant lung function compared with the patients with mild to moderate lung function.Oxidative stress and increased NLRP3 mRNA expression were associated with exacerbation of COPD.3.There was a positive correlation between the serum MDA level and NLRP3 mRNA expression in COPD patients.The serum SOD level was negatively correlated with NLRP3 mRNA expression.FEV1%pred was negatively correlated with NLRP3 mRNA expression.Oxidative stress mediates the activation of NLRP3 inflammatory corpuscule and affects the progression of COPD.
