Objective: To explore the expression of Rac1,Rac2 and Rac3 subtypes in peripheral blood mononuclear cells of patients with CHD and their correlation with NLRP3 inflammatory corpuscles.Methods: Patients admitted to the department of cardiology of the second hospital of Da Lian medical university from June 2018 to January 2019 due to chest pain were collected.According to the inclusion and exclusion criteria,8 healthy Control patients,18 Stable Angina(SA)patients,20 Unstable Angina(UA)patients and 16 Acute Myocardial Infarction(AMI)patients were selected.All subjects were asked about their medical history in detail by cardiologists,including age,gender,height,weight,smoking history,history of hypertension and history of diabetes.Venous blood of the selected patients was collected from the hollow elbow by the cardiology nurses from the morning after admission,and blood routine,liver biochemistry,renal function,blood lipids,myocardial markers,thyroid function and so on were measured.Two additional blood samples of EDTA anticoagulant tubes were taken,with a volume of about 6ml.Mononuclear cells were extracted by Ficoll density gradient separation.The expressions of Rac1,Rac2,Rac3,NLRP3,caspase-1,and IL-1 ? and IL-18 were detected by western blot,and the expressions of each subtype of Rac,NLRP and its downstream inflammatory factors were determined.Coronary angiography was performed by experienced physicians in the department of cardiology,and Gensini score was used to evaluate coronary artery lesions.SPSS 22 software was used for statistical analysis of relevant data.Results: 1.Compared with the control group,Rac1 expression in the stable angina,unstable angina and acute myocardial infarction groups increased successively,with statistical significance(P < 0.001).Rac2 expression was significantly increased in stable angina,unstable angina and acute myocardial infarction groups(P < 0.001).Rac3 expression decreased successively in stable angina pectoris,unstable angina pectoris,and acute myocardial infarction group,with statistical significance(P <0.01).2.Expressions of NLRP3,caspase-1,IL-1 ? and IL-18 in healthy control group,stable angina,unstable angina and acute myocardial infarction group increased successively;The expression of pro-caspase-1 in stable angina pectoris was higher than that in unstable angina pectoris and acute myocardial infarction group.3.Rac1 protein expression was positively correlated with the protein expression of NLRP3 and its downstream factor IL-1?.Rac2 protein expression was positively correlated with NLRP3 and its downstream factor IL-1 ?.Rac3 protein expression was negatively correlated with NLRP3 and its downstream factor IL-1?.Conclusion: 1.In monocytes with different types of coronary heart disease,Rac1 increased with the severity of CHD;2.Rac2 expression was significantly increased with the increase of severity of CHD.3.Rac3 expression decreased as the severity of cad increased.4.Rac1 was positively correlated with the protein expression of NLRP3 and its downstream inflammatory factor IL-1 ?.Rac2 was positively correlated with the protein expression of NLRP3 and its downstream inflammatory factor IL-1?.Rac3 was negatively correlated with the protein expression of NLRP3 and its downstream inflammatory factor IL-1?.
