Objectives: To preliminarily investigate the effect and mechanism of microRNA-608 on the invasion of SGC7901 cells and provide experimental data for revealing the molecular mechanism of gastric cancer.Methods: 1.The microRNAs binding RTN4 gene were predicted and analyzed by bioinformatics software.The expression of microRNA-608 was detected by real-time fluorescent quantitative PCR in gastric cancer SGC7901 cells.2.The 3'-UTR region binding site of RTN4 Gene Regulated by microRNA-608 was predicted by bioinformatics software.Targeted binding of between microRNA-608 and RTN4 gene 3'-UTR was detected by Luciferase reporter gene.3.The expression of RTN4 was detected in MGC803 cells transfected into microRNA-608.The effect of microRNA-608 on migration and invasion of SGC7901 cells was observed by scratch test and Transwell migration and invasion test.4.The activation of NF-kappaB signaling pathway was detected by Western-blotting in SGC7901 cells.Results:1.The bioinformatics software predicted the RTN4 gene binding by microRNA-608.The results of real-time fluorescence quantitative PCR showed that the expression of microRNA-608 was low in SGC7901 cells(P<0.05).2.microRNA-608 binds to 591-598 nucleotides of RTN4 gene3'UTR.The luciferin reporter gene system activity of 3'UTR sequence containing RTN4 gene was decreased after the overexpression of microRNA-608,and the expression of RTN4 gene and protein was decreased in SGC7901 cells(P<0.05).3.The scratch assay showed that the migration distance of SGC7901 cells was small and the migration ability of SGC7901 cells was decreased after the overexpression of microRNA-608(P<0.05).The results of Transwell migration and invasion experiment showed that the number of migration and invasion cells and the ability of migration and invasion of SGC7901 cells was decreased significantly after the overexpression of microRNA-608(P<0.05).4.The results of Western-blotting showed that the overexpression of microRNA-608 inhibited the activation of NF-kappaB signaling pathway in SGC7901 cells(P<0.05).Conclusions: microRNA-608 down-regulates the expression of RTN4,inhibits the activation of NF-kappaB pathway and reduces the migration and invasion ability of SGC7901 cells.
