Objective As a common cancer,gastric cancer has been a serious threat to human health.Along with the research progress of gastric cancer gene level,studies have shown that neuropilin 1 plays an important role in the occurrence and development of gastric cancer.This study used The Cancer Genome Atlas database to analyze the expression of NRP-1 in gastric cancer tissues and its correlation with the prognosis of patients.At the same time,the gastric cancer tissues of our hospital were used to verify the expression of NRP-1 and its relationship between pathological features and prognosis.Methods The gastric cancer-related data were downloaded from the TCGA database,and the differentially expressed genes in gastric cancer tissues compared with the adjacent tissues were analyzed.It was analyzed whether the differential expression of NRP-1 in the database and whether its differential expression was related to the prognosis of patients;The clinicopathological data of 109 patients with gastric cancer who underwent surgical treatment in our department from April 2009 to April 2010 were retrospectively analyzed.The expression of NRP-1 in gastric cancer and adjacent tissues was detected by immunohistochemistry.The relationship between NRP-1 and the clinicopathological features and prognosis of gastric cancer was analyzed.Results Based on the analysis of gastric cancer data in the TCGA database,NRP-1 was highly expressed in gastric cancer compared with normal tissues(18.19:6.65)and the expression of NRP-1 was correlated with the prognosis of patients(P<0.05).In 109 patients,61 patients had high expression of NRP-1 in gastric carcinoma,and 48 cases had low expression.In normal tissues,11 cases were highly expressed and 98 cases were low expression,and the difference between the two groups was statistically significant(P<0.05).From the statistical analysis,the expression of NRP-1 in gastric cancer tissues was not significantly different in ageand gender(P>0.05);the expression rate of NRP-1 in gastric cancer tissues with tumor diameter greater than or equal to 50 mm was significantly increased(P<0.05).The expression of NRP-1 between the pathological stage pT1/T2 and pT3/T4 group was statistically significant(P<0.05),and its expression increased with the increase of stage;The expression of NRP-1 increased significantly with the increase of lymph node positive number(P<0.05).Moreover,the histologically,the high expression rate of NRP-1 was significantly increased in the poorly differentiated or undifferentiated group(P<0.05);The high expression of NRP-1 was associated with tumor diameter,primary tumor status,lymph node metastasis,lymph node metastasis,and histological differentiation(P<0.05).COX regression(proportional risk model)multivariate analysis showed that NRP1 expression was an independent risk factor for the prognosis of gastric cancer patients(P<0.05).Conclusion NRP-1 was differentially expressed in gastric cancer tissues by TCGA database analysis,and this difference in expression was correlated with prognosis.Clinically,it is concluded that NRP-1 promotes tumors in the progression of gastric cancer,and its high expression is associated with the malignant biological behavior of gastric cancer;the high expression of NRP-1 is negatively correlated with the survival rate of gastric cancer patients.NRP-1 may be used as a tumor marker for predicting the prognosis of patients with gastric cancer.
