Objective:Despite the great progress in the treatment of gliomas in surgical, radiotherapy, and chemotherapy, there is still insufficient evidence-based medical evidence for immunotherapy of gliomas. So,the current meta-analysis were conducted to assess survival and clinical response of immunotherapy in patients with glioma. Methods:We systematically searched PubMed,the Cochrane Library,Medline,EMBASE,and Web of Science for relevant parallel randomized controlled trials(RCTs)and properly controlled non-randomized studies(NRS),quasi-randomized controlled trials(quasi-RCTs),and controlled clinical trials with historical control groups investigating the clinical efficacy of immunotherapy in patients with glioma, the search period from inception to January 2019.The obtained literature was carefully matched and screened, and valid data was extracted.Meta-analysis was performed using Review manager 5.3 software to evaluate the effect of immunotherapy on the survival rate of patients with malignant glioma at various stages and its subgroup(active immunotherapy group and passive immunotherapy group)for analysis. Results:Of 1415 suirapy group table studies,20 studies(four randomized controlled trials and sixteen non-randomized studies)involving 1607 patients were finally included.Compared to the control the(clinical trials group),the immunotherapy group showed better over survival without serious adverse events.One-year overall survival was significantly higher in immunotherapy groups[odds ratio(OR)=2.48;95%CI=1.74-3.54;P<0.00001].Two-year overall survival was significantly higher in immunotherapy[odds ratio(OR)=3.30;95%CI=2.24-4.87;P<0.00001].Three-year overall survival was significantly higher in immunotherapy as well[odds ratio(OR)=5.84;95%CI=3.15-10.83;P<0.00001].Subgroup analysis showed that trials designed as immunotherapy group obtained better results in the active immunotherapy group in this study. Conclusion:Immunotherapy vaccine can significantly improve over survival, with better results in the active immunotherapy group,of glioma patients.
