| Objective:To study the effect of 10%nitric acid decalcification on EGFR mutation rate in lung adenocarcinoma bone metastasis specimens in a short time,and to analyze the relationship between EGFR mutations and clinical features of lung adenocarcinoma patientsMethods:1.A total of 77 patients with bone metastases from lung cancer who were treated in our hospital from June 2013 to August 2018.The pathological types were confirmed as adenocarcinoma by comprehensive analysis of morphology,immunohistochemistry and clinical imaging data.Among them,32 specimens were treated with 10%nitric acid as the experimental group,and 45 specimens were not treated with 10%nitric acid as the control group;2.EGFR mutation detection was performed by ARMS-PCR;3.SPSS 22.0 was used for statistical analysis of all data,and chi-square test and Fisher’s exact probability method were used for analysis and comparison.Results:1.The overall mutation rate of EGFR in the experimental group and the control group was 54.55%(42/77);2.The EGFR mutation rate in the experimental group was 50.00%(16/32),while that in the control group was 57.78%(26/45),P=0.499>0.05,and the difference was not statistically significant;3.The mutation rate of EGFR in male patients was 47.62%,and that in female patients was 62.86%,P=0.181>0.05,the difference was not statistically significant;4.The EGFR mutation rate of patients older than 50 years old was 53.62%,and that of patients younger than 50 years old was 62.50%,P=0.721.The difference was not statistically significant;5.The EGFR mutation rate of smoking patients was 33.33%,and that of non-smoking patients was 65.12%,P=0.032<0.05,and the difference was statistically significant;6.There were 15 cases of mutation in 19-del,accounting for 35.72%of all 42 cases,and there were 24 cases of L858R mutation,accounting for 57.14%of all mutations.Both of them accounted for 92.86%of all mutationsConclusions:1.For small specimens of puncture,if the time of decalcification is controlled within 10-30min,the use of 10%nitric acid decalcification in the treatment of lung adenocarcinoma bone metastasis specimens has no effect on subsequent immunohistochemical detection,and EGFR mutation detection by ARMS-PCR.;2.In this study,EGFR mutations were more common in non-smoking patients;But,there was no difference in gender or age;3.Common mutations in EGFR mutations include 19-del and L858R,accounting for more than 90%of all mutations;Among them,L858R were more common in bone metastatic specimens of lung adenocarcinoma in our study. |
