Objective: This study intends to predict LincRNA associated with gastric cancer(GC)and its related biological signaling pathways,and to screen specific LincRNA as a potential biomarker for GC prognosis.Methods: Firstly,the RNA-seq data of 375 GC tissues and 32 paracancer from GC patients were loaded from The Cancer Genome Atlas(TCGA)database,and the relevant clinical information of GC patients was obtained,with the LincRNA and mRNA extracted,respectively.Secondly,the differentially expressed LincRNA and mRNA were screened using the Bioconductor package edgeR.Thirdly,the Weighted Gene Co-expression Network Analysis(WGCNA)was applied to analysis the data,thus obtaining the module and corresponding clinical features with the GC.The central gene was found by CytoHubba plugin to get the key LincRNA which was the most related to GC occurrence.The correlation between the key LincRNA and the LincRNA as well as mRNA,which were associated with GC occurrence and metastasis was explored through target gene prediction and the biological function analysis.Hence,theco-expression network of LincRNA and mRNA was constructed by Cytoscape software.The Kaplan-Meier software was exhibited to explore the relationship between LincRNA and overall survival rate of GC patients.Results: Firstly,2200 differentially expressed LincRNAs and 4623 differentially expressed mRNAs were filtered in this study.This study found that MEbrown module and MEturquoise module had the greatest correlation with tumor grade.Five central genes(Hub gene)in the LincRNA module,with the greatest correlation with GC,were screened out,including BARX1-AS1,CARMN,FENDRR,GAS1 RR and LINC01354.Also,this study found there were seven major KEGG pathways,which were MAPK Signaling Pathway ? CELL CYCLE Signaling Pathway ? P53 Signaling Pathway ? P13K-AKT Signaling Pathway?WNT Signaling Pathway?TGF-BETA Signaling Pathway?ADHERENS JUNCTION Signaling Pathway related to GC pathogenesis.In addition,LincRNA and RNA networks were constructed to analyze and verify Hub genes.The survival rate of the five high-expression LincRNA group was lower than that of the low-expression group.Conclusion: This study suggests that BARX1-AS1,CARMN,FENDRR,GAS1 RR and LINC01354 can be used as potential biomarkers for GC prognosis.
