Objective: To test the VEGF-C protein in nasopharyngeal tumor tissues and the expression of VEGF-C influence on human lymphatic endothelial cell biology behavior,analytical expression of VEGF-C relation with clinical pathological characteristics of nasopharyngeal carcinoma(NPC),explore the mechanism of lymphatic metastasis of nasopharyngeal carcinoma(NPC),looking for a new index for the prognosis of nasopharyngeal carcinoma(NPC) assessment and tries to explore the resistance to lymphatic metastasis of nasopharyngeal carcinoma(NPC) treatment.Methods:Part ?:1)Collecting and grouping the subjects according to the conditions of the experiment;2)Detecting the expression of VEGF-C protein by immunohistochemical staining(SP);3)Analyzing the experimental results by statistical software SPSS19.0.Part ?: 1)Cultured human lymphatic endothelial cells in vitro;2)Plasmid transfection by electroporation;3)The subjects were divided into three groups: untreated human lymphatic endothelial cells group,transfected empty plasmid pci-neo human lymphatic endothelial cells group and transfected recombinant plasmid pci-neo-anti-vegf-c human lymphatic endothelial cells group;3)Western blot method was used to detect vegf-c expression in lymphatic endothelial cells of the three groups.4)MTT method,cell scratch test,cell invasion test and flow cytometry were used to detect the proliferation,migration,invasion and apoptosis of lymphatic endothelial cells in the three groups.5)Statistical software SPSS19.0 was used for statistical analysis of the experimental results.Results: Part ?: 1)The positive rate of VEGF-C protein expression in nasopharyngeal cancer tumor tissues was higher than that in normal nasopharyngeal mucosa tissues,and the difference was statistically significant(P<0.05);2)The positive expression rate of VEGF-C protein in nasopharyngeal cancer tumor tissues of the group with lymph node metastasis was significantly higher than that of the group without lymph node metastasis,and the difference was statistically significant(P<0.05);3)The positive expression rate of VEGF-C protein in tumor tissues of stage ? nasopharyngeal carcinoma was higher than that of stage ? and ? nasopharyngeal carcinoma,and the difference was statistically significant(P<0.05);4)The positive expression rate of VEGF-C in the tumor tissues of non-keratinized and undifferentiated carcinoma was higher than that of non-keratinized and differentiated carcinoma,and the difference was statistically significant(P<0.05).There was no statistically significant difference in the positive expression rate of VEGF-C between keratinized and non-keratinized carcinoma(P>0.05).Part ?:1)VEGF-C protein was expressed in all three groups of human lymphatic endothelial cells,but the expression level was different.The expression level of VEGF-C protein in human lymphatic endothelial cell line transfected with recombinant plasmid PCI-neo-anti-VEGF-C was significantly higher than that in blank plasmid PCI-neo human lymphatic endothelial cell line and untreated human lymphatic endothelial cell line.The difference was statistically significant(P < 0.05),but there was no significant difference between the latter two groups in the content of VEGF-C in human lymphatic endothelial cells(P > 0.05).2)Transfection recombinant plasmid PCI-neo-anti-VEGF-C group of lymphatic endothelial cells proliferation,migration and invasion ability were higher than no-load plasmid transfection PCI-neo group and untreated group of lymphatic endothelial cells,the difference was statistically significant(P < 0.05),and the proliferation,migration and invasion abilities of lymphatic endothelial cells in the latter two groups were not statistically significant(P>0.05);3)The apoptosis rate of human lymphatic endothelial cells transfected with recombinant plasmid PCI-neo-anti-VEGF-C was lower than that of no-load plasmid transfection PCI-neo and untreated human lymphatic endothelial cells.The difference was statistically significant(P < 0.05),but there was no significant difference in apoptosis rate of human lymphatic endothelial cells between the latter two groups(P > 0.05).Conclusions:1)The expression of VEGF-C is correlated with lymph node metastasis,clinical stage and differentiation degree of nasopharyngeal carcinoma,providing a new reference basis and index for the treatment and prognosis evaluation of nasopharyngeal carcinoma.2)VEGF-C may promote lymphatic metastasis of nasopharyngeal carcinoma by enhancing the proliferation,migration and invasion of human lymphatic endothelial cells and inhibiting their apoptosis.
