Objective:Guided by the theory of traditional Chinese medicine and combined with clinical manifestations,to evaluate the effects of vinegar-baked Bupleuri Radix with soothing liver function and liver toxicity by means of the traditional Chinese medicine literature mining combined with modern pharmacology and toxicology research.To provide strong experimental data support for the safety and effectiveness of late-stage clinical medication.Methods:Combination of traditional Chinese medicine syndrome points and sucrose preference test and testing the neuroendocrine immune network in sensitive indicators,which include NE,5-HT,ACTH,CORT,TNF-?,IL-6,IL-1?and hepatotoxicity of sensitive indicators which include ALT,AST,TBIL,ALP.And calculate each rat liver,brain,thymus and splenic of dirty body or brain ratio.In vitro L02 cell assay was used to further reveal the toxic dose changes of water extract components of bupleurum chinensis.Results:Compared with normal control group,the model group rats'fur color was sparse and non-slippery,with abnormal excreta,drooping eyelids and prone idle movement,and the consumption of sugar water are reduced.The activity of NE,5-HT,IL-1?are decreased,on the contrary,the content of ACTH,CORT,TNF-?,IL-6 all increased significantly.The water extract components of bupleurum vinegar could improve the symptoms of model rats.In addition,the treatment effect of the high and medium dose groups of water extract components of bupleurum chinensis prepared with vinegar was better than that of the high dose group of water extract components of bupleurum chinensis prepared with vinegar.The indexes of ALT,AST,ALP and TBIL were detected,and the indexes of high and medium water extract components of bupleurum chinensis were significantly higher than those of high and medium water extract components of bupleurum chinensis.In vitro,the activity of water extracts from bupleurum chinense decreased from 96.03% to 11.26%,and the IC50 of cells were 1173.87 mg·L-1 and 1205.59 mg·L-1,respectively.Conclusions:The water extract components of bupleurum vinegar before and after the preparation of bupleurum vinegar improved the symptoms of the model rats,and the vinegar preparation of bupleurum was more effective in improving the symptoms of the rats.The rats of NE,5-HT,IL-1?concentration increases,ACTH,CORT,TNF-?,IL-6concentration decreased.The toxic and side effect of the group with high and medium water extracts of bupleurum chinensis was stronger than that of the group with high and medium water extracts of bupleurum chinensis.At a cellular level,The water extract of bupleurum chinensis made of vinegar can cause liver damage,and the damage intensity is higher than that of water extract of bupleurum chinensis made of vinegar.The use of vinegar in the clinical application of bupleurum chinensis should follow the doctor's advice.
