Study On The Compatibility Of Anti-depressant Prescription Containing Radix Bupleuri And Radix Scutellariae

The prescriptions containing Radix Bupleuri and Radix Scutellariae are common prescriptions in clinical treatment of depression.This paper collected a large sample of reports on the treatment of depression containing Radix Bupleuri and Radix Scutellariae,and using data mining methods,studied its clinical common syndromes and syndromes,drug compatibility rules,the dosage of Radix Bupleuri and Radix Scutellariae membranaceus and their compatibility ratio.Using animal experiments,the effects of Radix Bupleuri,Radix Scutellariae and their compatibility on depression model were observed.In order to preliminarily clarify the rule of treatment of this class of prescriptions and the scientific connotation of the compatibility of the prescriptions,and it provides a basis for the clinical application of prescriptions containing Radix Bupleuri and Radix Scutellariae in the treatment of depression.The main content of the thesis is divided into two parts:data mining and experimental research.MethodsFirst,Data mining Collect a large sample of reports on the treatment of depression in the past 36 years containing Radix Bupleuri and Radix Scutellariae.According to "Chinese Pharmacy"and"Pharmacopoeia of the People's Republic of China",standardize the names and categories of drug,and establish a database of prescription information.SPSS software was used for frequency analysis,association rules and cluster analysis to explore the drug rule and prescription principle of anti-depressant prescription.Second,Experimental Study The rat depression model was prepared by CUMS.The rats were randomly divided into blank control group,model group(CUMS),fluoxetine group,Radix Bupleuri group,Radix Scutellariae group,Radix Bupleuri-Radix Scutellariae low-dose group and Radix Bupleuri-Radix Scutellariae high-dose group.Each administration group was continuously administered for 3 weeks,and its behavioral change was observed.ResultsFirst,the data mining part1The 207 large sample documents collected in this study involved 8553 cases of medical records and 145 flavors of drugs.2The syndromes treated were mostly liver stagnation and qi stagnation,.It's frequency is 1169 times and frequency is 33.0%,which is much higher than other syndrome types;Among them,there were liver stagnation phlegm obstruction type(9.6%),liver depression phlegm heat type(9.2%),liver depression and spleen deficiency type(6.3%),liver stagnation blood stasis type(5.3%),etc.The main disease position of the syndromes treated by such prescriptions is mostly liver,which is related to heart,gallbladder,spleen and kidney.Its syndrome(sickness)can be divided into tw^o ends.The empirical evidence is qi stagnation,fiery,stagnation,blood stasis,dampness.The deficiency syndrome is qi deficiency,blood deficiency,yin deficiency,and yang deficiency.Among them,the qi stagnation of each syndrome is the first of all syndromes.It's frequency is 86.9%.It indicates that qi stagnation is the main pathogenesis of the disease treated by prescriptions containing Radix Bupleuri and Radix Scutellariae.Secondly,the frequency of heat and phlegm depression were 26.2%.The third is qi deficiency with frequency of 11.6%.3The frequency statistics of single-flavor drugs showed that there are 31 kinds of common drugs compatible with Radix Bupleuri and Radix Scutellariae.Among them,the frequency of Pinellia is the highest,with a frequency of 80.3%.,far exceeding other drugs.Licorice and poria cocos were next,with frequency of 56.7%and 54.2%,respectively.The compatible drugs were classified according to their efficacy,and the main drugs were sedative drugs,phlegm and invigorating medicine.Among them,the medicine for calming the nerves is mainly based on the role of calming the nerves in the town.It is commonly used in keels and oysters.Drugs that nourish the heart and soothe the nerves such as Acorus tatarinowii,Radix polygala,zizyphus jujube kernel,albizzia bark,acacia stem,etc;Pinellia ternata and bamboo shavings are commonly used in phlegm drugs;Ginseng drugs(ginseng,codonopsis pilosula,Radix pseudostellariae)are commonly used in phlegm invigorating medicine.4Through correlation analysis,the combination of the two drugs containing Radix Bupleuri and Radix Scutellariae prescription has the highest confidence level:bamboo shavings?pinellia,up to 100%;the highest support is poria cocos?pinellia(49.5%);the first with higher support and confidence is oysters?Keel(96.3%,42.0%).These drugs are almost composed of or contain the drugs contained in Bupleurum and keel oyster soup.For the drug without this side,the highest confidence is angelica?Radix paeoniae alba medicine(76.1%),and the highest support is stone calamus?turmeric(22.7%).The three commonly related drugs are keel,poria cocos?pinellia?oyster,poria cocos?pinellia,cassia twig,poria cocos?pinellia.The four commonly related drugs are oysters,keels,and scorpions?pinellia.5Through cluster analysis,four groups of drugs are obtained,namely Cl keel,oyster,cassia twig,rhubarb,codonopsis,poria,ginger,jujube,pinellia,which are the composition of bupleurum plus keel oyster soup;C2 god eight,salvia,acacia,suanzaoren,and these 4 tastes all have nourishing and calming effect;C3 lily,atractylodes,clam shell,rhizoma cyperi,rhizoma ligustici wallichii,gardenia,angelica,and this group of 7-flavored medicine has the functions of nourishing yin,nourishing blood,strengthening spleen,soothing liver,clearing heat and promoting blood circulation;C4 tamping,bamboo shavings,tangpi,polygala,and this group of 4 herbs has the effect of clearing away heat and removing phlegm.6According to the statistics on the amount of Radix Bupleuri and Radix Scutellariae,the minimum dosage of bupleurum is 6g.The maximum dosage is 30g.The average dosage is 12.5±3.7g?and the usual dosage is 10g,12g and 15g;The minimum dosage of Astragalus is 3g.The maximum dosage is 18g.The average dosage is 10.2±2.2g,and the commonly used doses are lOg and 12g.There are as many as 18 kinds of ratio for Radix Bupleuri and Radix Scutellariae used in clinical practice,of which 1:1 is the most.The frequency is 2982,accounting for 37.3%of all cases.Followed by 3:2,and the frequency is 1583,accounting for 19.8%.Second,the experimental research partThe Radix Bupleuri group,Radix Scutellariae group and Radix Bupleuri-Radix Scutellariae high and low dose groups can increase the sucrose preference rate of CUMS rats,significantly increasing the number of penetrating,standing and wiping the face of CUMS rats,and shortening the time of forced swimming suspension in CUMS rats.In terms of succulent water preference and autonomic activity in CUMS rats,the effect of Radix Bupleuri-Radix Scutellariae high and low-dose groups was higher than that of Radix Bupleuri and Radix Scutellariae,while the suspension time of forced swimming in CUMS rats was higher than that of Radix Bupleuri and Radix Scutellariae group.ConclusionFirst,the data mining part1The pathogenesis characteristics with the syndrome of Radix Bupleuri and Radix Scutellariae has empirical evidence and deficiency.Based on empirical evidence,there are false and real mixed.Pathogenic factors are mainly qi stagnation,and there are changes in fire,oysters,etc.Deficiency syndrome is less,qi deficiency,blood loss,yin injury and other performance.2The combination of Radix Bupleuri and Radix Scutellariae is the core of the prescriptions containing Radix Bupleuri.Radix Bupleuri and Scutellaria scutellaria were combined:On the one hand,the lifting and dispersing of Radix Bupleuri and the lowering and dispersing of Radix Scutellariae can help to mediate the cardinal organ,adjust the rising and falling of qi and enhance the function of relieving depression;On the other hand,Radix Bupleuri can relieve liver depression,and Radix Radix Scutellariae can clear away liver and gallbladder fire,to reduce the effect of liver and heat.For those with qi depression but no obvious heat phenomenon,the Radix Scutellariae can be used to prevent them from turning into heat.3Clinical application of Radix Bupleuri and Radix Radix Scutellariae decoction for the treatment of depression,Radix Bupleuri and Radix Radix Scutellariae take a moderate dose;and Radix Bupleuri and Radix Radix of this kind of prescription.Second,the experimental research partAnimal experiments showed that the Radix Bupleuri group,Radix Scutellariae group and Radix Bupleuri-Radix Scutellariae high and low dose groups had good antidepressant effects,and compatibility of Radix Bupleuri-Radix Scutellariae had better antidepressant effects than single drugs.
