Objective:1.Detection of the difference in Sap2 activity between Candida albicans resistant strains and susceptible strains.2.To explore the virulence and pathogenicity in resistant strains and sensitive strains of Candida albicans by animal(mice)experiments.3.To detect the expression levels of Sap2 and ERG11 mRNA of Candida albican sensitive strains and drug resistant strains by RT-PCR and to explore their relationship with Candida albicans resistance.Methods:1.Take all the resistance to itraconazole,fluconazole and voriconazole identified by the laboratory through drug sensitivity test,only resistant to itraconazole,only resistant to fluconazole,ten strains were completely sensitive to fluconazole,itraconazole and voriconazole,and the Sap activity of the 50 bacterium was detected by BSA medium,and statistical analysis was performed.2.the experimental strains were injected into the tail vein of the experimental mice,and the number of deaths was recorded,and statistical analysis was performed.3.The relative expression levels of Sap2 and ERG11 mRNA were detected by RT-PCR,and the relationship between drug resistance and the expression level was analyzed.Results:1,Sap activity test results: fluconazole,itraconazole and voriconazole cross-resistant strains(10 strains),fluconazole resistant strains(10 strains),itraconazole resistant strains(10 strains),voriconazole The drug-resistant strains(10 strains),all three sensitive strains(10 strains),a total of 50 Candida albicans have transparent rings.Fluconazole-resistant group,Pz value=0.631±0.010;Itraconazole-resistant group,Pz=0.701±0.028;voriconazole-resistant group Pz=0.623±0.011;cross-resistance group,Pz=0.590± 0.023;sensitive group,Pz value=0.795±0.016;the difference of Sap activity between the cross-resistance group and other strain groups was statistically significant;the Sap activity of the sensitive strain group was lower than that of the resistant strain group,and the difference was statistically significant(F= 18.693,P < 0.001).2.Virulent pathogenicity results of mouse artificial bacteremia model: fluconazole resistant group,itraconazole resistant group voriconazole resistant group,cross-resistant group,sensitive group,blank group The survival time was 22 days,18 days,13.5 days,12 days,20 days and 30 days.The Log-rank test showed that the difference in survival time between the different groups was statistically significant(=25.62,P=0.00),that is,the pathogenicity and virulence of the resistant group were significantly higher than those of the sensitive group.3,SAP2,ERG11 mRNA expression detection results: the cross-resistance group SAP2 mRNA relative expression was(6.9127 ± 1.57)significantly higher than the sensitive strain group(2.4418 ± 0.91)(P <0.05),the difference was statistically significant The relative expression of SAP2 mRNA in the cross-resistance group was higher than that in the single-drug resistant strain(the relative expression of SAP2 mRNA in the itraconazole-resistant group was 5.3164±0.56,and the relative expression of SAP2 mRNA in the fluconazole-resistant group was 5.2164±0.74.The relative expression of SAP2 mRNA in the voriconazole-resistant group was 5.5927±0.57,and the difference was statistically significant.The relative expression of ERG11 mRNA in thecross-resistance group was significantly higher than that in the sensitive strain group(1.89±1.89±).0.93)(P<0.05),the difference was statistically significant;the expression of cross-resistant strains was higher than that of the three groups of single-resistant strains(the relative expression of ERG11 mRNA in the fluconazole-resistant group was5.263±0.43,Yiqu The relative expression of ERG11 mRNA in the Conazole-resistant group was 5.311±0.67,and the relative expression of ERG11 mRNA in the voriconazole-resistant group was 6.022±0.29).The difference between the fluconazole-resistant group and the itraconazole-resistant group was statistically significant.Academic significance,with Vulcan There was no significant difference between the drug.Conclusion:1.Among the resistant strains of Candida albicans,the activity of Sap2 is increased,and the pathogenicity of virulence is enhanced.2,SAP2,ERG11 high expression and Candida albicans cross-resistance,single resistance has a certain relationship.3.Candida albicans SAP2 and ERG11 are positively correlated in azole resistance,and there may be positive regulation between the two genes.
