Objective: To explore the effect of the NEAT1 on the proliferation,invasion,migration and apoptosis of the prostate cancer cell(DU145)and the molecular mechanism.Methods: The expression level of lnc-NEAT1 and miR-128-3p in the prostate cancer and the adjacent tissues was measured by qRT-PCR.Next,we test the expression level of lnc-NEAT1 and miR-128-3p in prostate cancer cells and normal prostate epithelial cells.Western blotting was used to detect the expression level of ZEB1.CCK-8 assay was used to detect the proliferation activity of cells.Transwell assay was used to detect the invasion and migration of cells.Annexin V-FITC /PI double staining and flow cytometry were used to detect the apoptosis rate.The dual-luciferase reporting system is used to detect the targeted interaction between lncRNA-NEAT1 with miR-128-3p and miR-128-3p with ZEB1.Results: The lncRNA-NEAT1 expression level in the prostate cancer tissue is significantly higher than that in the adjacent tissues.While the expression level of miR-128-3p in prostate cancer tissues was significantly lower than that in adjacent tissues.Cell level test results show that the expression level of lncRNA-NEAT1 in the three prostate cancer cells is significantly higher than that of the normal prostate epithelial cells.The expression level of lncRNA-NEAT1 in DU145 is the highest.The expression level of miR-128-3p in three strains of prostate cancer cells was significantly lower than that in normal prostate epithelial cells.Among them,the expression level of miR-128-3p in DU145 was the lowest.Compared with the control group,over expression of lncRNA-NEAT1 significantly improve the proliferation,invasion and migration ability of DU145 cells,and the cell apoptosis rate was significantly decrease.Knock down lncRNA-NEAT1 significantly inhibit the proliferation,invasion and migration abilities of DU145 cells,and the cell apoptosis rate was significantly increased.Over expression of miR-128-3p significantly inhibited proliferation,invasion and migration of DU145 cells,and significantly promoted apoptosis.Knock down of miR-128-3p significantly promoted proliferation,invasion and migration of DU145 cells,and significantly reduced apoptosis.Over expression of ZEB1 significantly promoted the proliferation,invasion and migration of DU145 cells,and the apoptosis rate was significantly reduced.Knock down of ZEB1 significantly inhibited the proliferation,invasion and migration abilities of DU145 cells,and the apoptosis rate was significantly increased.The results of the dual-luciferase reporter gene test show that the lncRNA-NEAT1 is negatively regulate the miR-128-3p and miR-128-3p negatively regulates ZEB1.Compared with over expression or knockdown of miR-128-3p,the four groups of recovery experiments significantly restored the proliferation activity,invasion ability,migration ability and apoptosis rate of DU145 cells.Conclusion: The lncRNA-NEAT1 promote the proliferation,invasion,migration and reduce cell apoptosis of the DU145 cell by competitive binding to miR-128-3p with ZEB1.
