Objects: Long non-coding RNA Nuclear Paraspeckle Assembly Transcript 1(LncRNA NEAT1)was a novel lncRNA localized specifically to nuclear paraspeckles.Previous studies had shown that NEAT1 plays an important role in the pathogenesis of many human cancers through certain biological pathways.This study investigated the relationship between NEAT1 rs512715 and rs2239895 and susceptibility to lung cancer in Northeast of China.Methods: A case-control study was conducted to investigate the relationship between the two SNPs in NEAT1 and the susceptibility to lung cancer.The study contained 1021 subjects,including 462 lung cancer cases and 559 cancer-free controls.The basic information about of subjects was obtained by questionnaire survey,and 10 mL of peripheral blood was collected.The anticoagulant genomic DNA was extracted using the classic Phenol-Chloroform Method and genotyped using TaqMan? method.The data were sorted by Microsoft Excel 2013 and analyzed mainly by SPSS 20.0 statistical software.Differences in the distribution of age,gender,and cigarette smoking between cases and cancer-free controls were analyzed by student's t-test and chi-square test.At the same time,non-conditional logistic regression was used to calculate corrected OR and its 95% confidence interval(95% CI)to assess the risk between two SNPs and lung cancer.The crossover analysis was used to calculate the main effect and the interaction effect.The addition model and the multiplication model were used to explore the relationship between gene-environment interactions and lung cancer susceptibility.Results: No statistical association was found between the two SNPs of NEAT1 and lung cancer susceptibility.However,there was a statistical correlation between rs2239895 and that risk of lung squamous cell carcinoma by pathological type.Compared with wild homozygous genotype GG,heterozygous GC had a 0.805-fold increased risk of lung squamous cell carcinoma(OR=1.805,95% CI=1.168-2.789,P=0.008).For the dominant model,CC + GC genotypes increased the risk of lung squamous cell carcinoma(OR=1.668,95% CI=1.093-2.545,P=0.018).The crossover analysis results showed that compared with non-smokers carrying the protective genotype CC of rs512715,carrying risk genotypes GC and GG had a 7.235-fold higher risk of lung cancer(OR=8.235,95%CI=4.012-16.903,P<0.001).Rs2239895 had the similar results,OR=8.807,95% CI=2.878-26.953,P<0.001.However,the relationship between these two SNPs and the risk of lung cancer were not found in the stratified analysis,nor was the relationship between gene-environment(cigarette smoking)interaction and the incidence of lung cancer in the additive model and the multiplicative model.Conclusion: There were no statistical correlation between the two SNPs of NEAT1 and susceptibility to lung cancer.According to pathology type stratification analysis,the rs2239895 polymorphism of NEAT1 can increase the risk of lung squamous cell carcinoma.The interaction between the two SNPs and smoking status had no significant effect on the risk of lung cancer.
