Background and ObjectiveGreat saphenous varicose veins are a common venous disorder characterized by degeneration vein valves,venous reflux,excessive venous dilation and tortusity and venous hypertension,and it could be complicated by venous rupture and bleeding,thrombosis,venous ulcer and superficial thrombophlebitis,but the underlying mechanism is unclear.Therefore,it has a potential clinical significance to investigate the differences in expression of matrix metalloproteinases(MMPs)and matrix metalloproteinase inhibitors(TIMPs)between normal and diseased splenic veins(DSVs)and varicose great saphenous vein(VGSVs)under high hemodynamics for illustrating the pathogenesis and targeted therapy of DSVs and VGSVs through analyzing the changing regularity of MMPs and TIMPs.MethodsFifteen specimens of DSVs from patients who underwent subtotal splenectomy were collected as DSVs group,and the control group involved another fifteen specimens of normal SVs from patients who suffered traumatic splenic rupture.Twenty-one specimens of VSGVs from patients who underwent high ligation and stripping plus transilluminated powered phlebectomy were collected as VSGVs group and the control group involved Twenty-one normal great saphenous veins.The mRNA and protein expression and the relative protein expression of MMP-2,MMP-9,TIMP-1,and TIMP-2 in the venous wall were observed and positive expression ratios were determined through RT-PCR and immunohistochemical and Western blot method respectively.ResultsClustered strong positive expression of MMP-2,MMP-9,TIMP-1,TIMP-2 proteins were observed in DSVs group and VGSVs group while negative results were found in SVs group and GSVs group.The positive mRNA and protein expression ratio and the relative protein expression of MMP-2,MMP-9,TIMP-1,TIMP-2 in DSVs group were significantly increased compared with that in SVs group,while in VGSVs group,thepositive mRNA and protein expression ratio and the relative protein expression of MMP-2,MMP-9,TIMP-1,TIMP-2 were significantly increased compared with that in GSVs group(all P < 0.05).Additionally,the VGSVs/GSVs ratio was significantly higher than the DSVs/SVs ratio(P < 0.05).However,there was no significant difference between the VGSVs/GSVs ratio and the DSV/SVs ratio as for the relative protein expression of MMP-2,MMP-9,TIMP-1,TIMP-2(P > 0.05).ConclusionUnder high hemodynamics,the expression of MMPs and TIMPs from splenic vein and great saphenous vein was in disequilibrium.These results may be one of the molecular mechanisms in vascular remodeling.
