Objective:This study aimed to investigate the relationship between DLK1(delta drosophilahomolog-like 1 or delta-like 1 homologue),RBM8A(RNA-binding motif protein 8A)and clinical data of patients with colorectal cancer(CRC)by examining the expression of DLK1 and RBM8A in CRC tissues and distant noncancerous tissues,and to analyze the correlation between DLK1 and RBM8A expressed in CRC.Methods:1The oncomine database and was used to screen out DLK1 and RBM8A abnormal expressed in CRC,UniHI database was used to found that DLK1and RBM8A have a relationship,and a literature review was conducted to determine the expressions of DLK1 and RBM8A in CRC tissues and their significance.2The experimental specimens were taken from 60 patients with CRC who underwent surgery from 2017 to 2018 in First Hospital of Qinhuangdao City.The data of patients with CRC were collected and analyzed.3Immunohistochemistry(qualitative)and western-blot(quantitative)assays were used to detect the expression of DLK1 and RBM8A in CRC tissues and their corresponding distant noncancerous tissues.4Statistical analysis was performed to analyze the relationship between DLK1,RBM8A and clinical data of CRC patients,and the correlation between expressions of DLK1 and RBM8A,if any,in patients with CRC.Results:The results of immunohistochemistry were as follows.1The expression of DLK1 and RBM8A in CRC tissues and distant noncancerous tissues.(1)The expression of DLK1 in CRC tissues was higher than that in distant noncancerous tissues,and there was statistically significant difference(χ~2=10.909,P=0.001).(2)The expression of RBM8A in CRC tissues was higher than that in distant noncanerous tissues,suggesting significant differences(χ~2=11.109,P=0.001).2The relationship between the expressions of DLK1 and RBM8A in CRC patients and clinical data.(1)The expression of DLK1 was correlated with tumor size(P<0.05),which was not correlated with gender,age,tumor differentiation,clinical stage,lymph node metastasis,serosal infiltration,distant metastasis,serum CEA level(P>0.05).(2)The expression of RBM8A was correlated with tumor size,degree of differentiation,and serosal infiltration(P<0.05),but was not correlated with gender,age,clinical stage,lymph node metastasis,distant metastasis,serum CEA level(P>0.05).3There was a relationship between the expressions of DLK1 and RBM8A in CRC tissues(r=0.431,P=0.001).The results of western-blot were as follows.1DLK1 was expressed in CRC tissues and distant noncancerous tissues,and the expression of DLK1 in CRC tissues was higher than that in distant noncancerous tissues,and there was statistically significant difference(t=2.391,P=0.027).2RBM8A was expressed in CRC tissues and distant noncancerous tissues,and the expression of RBM8A in colorectal cancer tissues was higher than that in distant noncancerous tissues,and there was statistically significant difference(t=2.225,P=0.038).Conclusions:1DLK1 is highly expressed in CRC tissues and is associated with tumor size in CRC patients,suggesting that it may be involved in cell proliferation of CRC.2RBM8A is highly expressed in CRC tissues and is associated with tumor differentiation,tumor size,and serosal infiltration in CRC patients,indicating that it may be involved in cell proliferation,invasion and malignancy of CRC.3The expressions of DLK1 and RBM8A in CRC may have a certain mutual regulation relationship,which lays the foundation for further research.
