| Objective:To explore the expression and clinical significance of Valosin-Containing Protein(VCP)in gliomas.Methods:1 Collection of clinical specimens:The samples of gliomas and normal brain tissues were collected from neurosurgery department of the second Hospital of Hebei Medical University for one year.According to the classification standards published by WHO in 2007,58 samples of human gliomas of grade I~Ⅳ were collected,including 26 for grade I~II,18 for grade III,14 for grade IV.At the same time,12 samples of normal brain tissue were taken as control group.2 Immunohistochemistry and Western Blot were used to detect and compare the expression of VCP protein in gliomas and normal brain tissues.Results:1 Immunohistochemistry showed that:There was no positive expression of VCP in normal brain tissue cells,but positive expression of VCP in glioma cells.With the increase of pathological grade of tumor,the expression of VCP increased gradually.The average optical density of normal brain tissue,grade Ⅰ~Ⅱ,grade Ⅲ and grade Ⅳ gliomas was(0.4275±0.0498)、(0.6910±0.1464)、(1.1840±0.2922)、(2.0520±0.3571),respectively.The difference of average optical density between any two groups was statistically significant(P<0.05).2 Western Blot results show:The expression of VCP in normal brain tissue cells was lower than that in glioma cells.The expression of VCP in glioma cells increased significantly,and the expression of VCP increased gradually with the increase of pathological grade of tumor.The grayscale ratio(VCP/β-action)of normal brain tissue,grade Ⅰ~Ⅱ,grade Ⅲ and grade Ⅳ gliomas was(0.2312±0.0171)、(0.4129±0.0130)、(1.0148±0.2047)、(2.1886±0.1538),respectively.The difference of gray values between any two groups was statistically significant(P< 0.05).Conclusions:1 The expression level of VCP in normal brain tissue was lower,and the expression level of VCP in glioma tissue was higher than that in normal brain tissue.2 The expression level of VCP in gliomas of different grades was different,and it showed an increasing trend with the increase of pathological grade of gliomas,which was closely related to the degree of malignancy of gliomas.VCP may be a new target for the treatment of glioma.The detection of VCP expression may be helpful to judge the prognosis of glioma patients and guide the clinical treatment. |
