Objective: Colorectal cancer is one of the common malignant tumors of the digestive system.Recurrence and metastasis after CRC treatment is one of the important factors that seriously affect the morbidity and mortality of CRC patients.Tumor recurrence and metastasis are closely related to tumor angiogenesis,and inhibition of tumor angiogenesis can effectively inhibit tumor recurrence and metastasis.Traditional Chinese medicine plays a good role in inhibiting tumor angiogenesis.This study was to observe the effect of Jiedu Huoxue Recipe on angiogenesis in colon cancer nude mice model and its targeted intervention effect on STAT3.Methods: Colorectal cancer nude mice model was established by inoculating colon cancer cells(HCT-116).50 nude mice were divided into model group,Chinese medicine high,medium and low dose groups,5-fluorouracil(5-FU)group.The model group was intragastrically administered with normal saline,high-,medium-,and low-dose Chinese medicines,and the 5-FU group was intraperitoneally injected with5-FU injection for 4 weeks.At the end of the fourth week,the nude mice were sacrificed,and the tumor inhibition rate was calculated.The microvessel density(CD34)was detected by immunohistochemistry,and the expression of VEGF and P-STAT3 was detected by Western Blotting.Results: The results of tumor inhibition rate of each group: high dose group of Chinese medicine: 58.78%;middle dose group of traditional Chinese medicine:47.29%;low dose group of Chinese medicine: 35.13%;group of 5-FU: 41.21%.The average tumor weight of the high,middle and low dose groups of Jiedu Huoxue Recipe was significantly lower than that of the model group(P<0.01),especially in the high dose group,and it showed a dose-effect relationship.Compared with the5-FU group,the average tumor weight of the high and middle dose groups of Jiedu Huoxue group was significantly lower(P<0.05).There was a statistically significant difference between the 5-FU group and the model group(P<0.01).It shows that Jiedu Huoxue has a good anti-tumor effect.The MVD of the high,middle and low dose groups of Jiedu Huoxue Recipe was significantly lower than that of the model group(p<0.01),indicating that Chinese medicine has obvious inhibitory effect on angiogenesis of CRC xenografts in nude mice,especially in the high dose group,and it is dose-effective.relationship.There was no significant difference in MVD between the 5-FU group and the model group(P>0.05).It shows that Jiedu Huoxue Recipe can effectively inhibit tumor angiogenesis.The expression of VEGF in the high,middle and low doses of Jiedu Huoxue Recipe was significantly lower than that in the model group(p<0.01),indicating that Jiedu Huoxue Recipe can significantly inhibit the expression of VEGF in CRC xenografts in nude mice,especially in the high dose group.A quantitative relationship.There was no significant difference in VEGF between the 5-FU group and the model group(P>0.05).It is indicated that Jiedu Huoxue Recipe can effectively inhibit the expression of VEGF in tumor.The expression of P-STAT3 in the high,middle and low dose groups of Jiedu Huoxue Recipe was significantly lower than that in the model group(p<0.01),indicating that Jiedu Huoxue Recipe can significantly inhibit the expression of P-STAT3 in CRC xenografts in nude mice,especially in the high dose group.Obvious,and in a dose-effect relationship.There was no significant difference in P-STAT3 between the5-FU group and the model group(P>0.05).It is indicated that Jiedu Huoxue Recipe can effectively inhibit the expression of tumor P-STAT3.Conclusion: Jiedu Huoxue Recipe has a good anti-tumor effect;Jiedu Huoxue Recipe can inhibit CRC angiogenesis;Jiedu Huoxue Recipe can inhibit STAT3 activation,thereby down-regulating VEGF expression and exerting its anti-tumor angiogenesis.
