Objective: This study was to explore the effect of platycodin D on PDGF-BB-induced proliferation of vascular smooth muscle cells(VSMCs)and its possible mechanism.Methods: Rat thoracic aorta vascular smooth muscle cells(A7r5 cells)were cultured in vitro.CCK-8 assay was used to evaluate the cell viability and proliferation inhibition of PDGFBB induced VSMCs induced by Platycodin D and MAPKs(U0126/SB203580/SP600125).Effect of Platycodin D on Cell Cycle Distribution of VSMCs Induced by PDGF-BB by Flow Cytometry.RT-PCR was used to detect m RNA expression levels of Cylin A,Cyclin B1,Cyclin D1,Cyclin E,CDK1,CDK2,and CDK4.Western-Blot was used to detect the protein expression levels of p-ERK1/2,ERK1/2,p-p38 MAPK,p38 MAPK,p-JNK and JNK.Results: 1.The effect of PDGF-BB on the proliferation of VSMCs: Compared with the control group,the viability of PDGF-BB was significantly increased at 20,30,and 40 ng / ml after 48,60 and 72h(p <0.05)ml group compared with 30,40 ng / ml group,no significant difference(P> 0.05).2.The effect of PD on proliferation of VSMCs induced by PDGF-BB: Compared with PDGF-BB group,the cell viability of PDGF-BB was significantly increased(p <0.05)at the concentration of 20,30,and 40 ng / ml after 36-60 hours of cell treatment.The inhibitory effect of platycodin D on proliferation of VSMCs induced by 20 ng / ml PDGF-BB was concentration-and time-dependent.Platycodin D could block the cells induced by PDGFBB in G1 / G0 and G2 / M phases with the corresponding m RNA expression of Cyclins and CDKs decreased.3.Platycodin D inhibits PDGF-BB-induced phosphorylation of ERK1/2,P38 and JNK in vascular smooth muscle cells.4.Effect of PD on proliferation of VSMCs induced by PDGF-BB and MAPK pathway: Compared with PDGF-BB 20ng/ml group,the cell viability was significantly decreased in the inhibitor group(U0126 / SB203580 / SP600125)20?M+ PDGF-BB 20ng/ml group(P <0.01),(U0126 / SB203580 / SP600125)20?M + platycodin D 4?g / ml + PDGF-BB 20 ng / ml group compared with(U0126 / SB203580 / 0.05)20?M + PDGF-BB 20 ng / ml group,no difference in cell viability was found between the two groups(P> 0.05).Conclusions: The inhibitory effect of platycodin D on the proliferation of VSMCs induced by PDGFBB may block both the G1/G0 phase and G2 / M phase cells by regulating the expression of cyclins and cyclin-dependent kinases,and ERK1/2,p38 MAPK,JNK dephosphorylation related.
