| Objective: To explore the mechanism of Enhance Qi Nourish Jing Formula in regulating angiogenesis of lung cancer by regulating apelin/APJ system.Methods: The lung cancer A549 and H1975 cells were treated with four groups such as Enhance Qi Nourish Jing Formula,apelin-13,the formula with apelin-13 and a control group.The ability of cell proliferation and migration were observed by CCK-8 kit,scratch assay and transwell test.The expression of RNA and protein of angiogenesis-related factors in each group was observed by RT-qPCR and Western Blot.The luciferase-expressing A549 cells were implanted into the lungs of mice to establish orthotopic models of lung cancer,The models were divided into six groups: the formula group,the apelin-13 group,the Cisplatin group,the formula with Cisplatin group,the formular with apelin-13 group and the control group,observing the changes of tumor by weight changes,organ indexes and bioluminescence images.RESULTS: Proliferation experiments showed that the proliferation of Enhance Qi Nourish Jing Formula group was significantly lower than that of the control group;In the invasion experiment,the number of cells in the Enhance Qi Nourish Jing Formula group was significantly less than that of the control group(p<0.05);RT-qPCR assay,in the Enhance Qi Nourish Jing Formula group,the expression of angiogenesis-related factors such as Apelin,APJ,HIF-1?,Smad3,and VEGF,was lower than that in the control group(p<0.05).Western blot assay showed that the expression of related factors(VEGF,HIF-1a,PI3K)was significantly down-regulated in the Enhance Qi Nourish Jing Formula group(p<0.05).In animal experiments,In the Cisplatin group and Enhance Qi Nourish Jing Formula+Cisplatin group,the weight of mice were higher than those in other groups;the organ indexes of A549 tumor-bearing mice treated with Enhance Qi Nourish Jing Formula+Cisplatin group was significantly lower than that of the control group(p<0.01);The fluorescence intensity of ROI of A549 tumor-bearing mice treated with Enhance Qi Nourish Jing Formula+Cisplatin group in the third week was lower than that in the second week.Conclusion: 1.Enhance Qi Nourish Jing Formula can antagonize the effect of apelin-13 on proliferation,migration and invasion of lung adenocarcinoma cells;2.Enhance Qi Nourish Jing Formula can antagonize the effect of apelin-13 on angiogenesis related factors VEGF,HIF-1a,PI3 K,down-regulating their expression;3.Enhance Qi Nourish Jing Formula plays a certain role in the survival of tumor-bearing mice;4.Enhance Qi Nourish Jing Formula combined with cisplatin on the A549 tumor-bearing mice have a certain Efficacy. |
