AIM: To observe the expression of apelin, ACE2, caveolin-1 of stroke-prone renovascular hypertensive rats' brain, heart, kidney and blood vessel tissues; the effect of intravenous injection of apelin on the blood pressure, heart rate and baroreflex sensitivity. Further, investigated the caveolin-1 signal pathway of apelin.Methods: The Stroke-prone renovascular hypertensive rats model was established. The expression of apelin was observed by immunochemistry and ELASA. The expression of ACE2 , p ERK1/2 and caveolin-1 were observed by Western Blot. Intravenous injection of apelin was followed by observation of the change in rat's blood pressure, heart rate and baroreflex sensitivity(the ratio of heart changes to blood changes).Results: The blood pressure of hypertensive group started rising at the end of 2th week , and reached the highest level at 4th week ,then stayed at the higher level. The ratios of heart to body weight of the hypertensive group were respectively 2.50 ± 0.16, 2.78 ± 0.17, 3.51±0.21, 3.94±0.22, 3.88 ± 0.34 at 0, 2, 4, 8, 12w. The ratios of heart to body of hypertensive group were somewhat higher than the sham operation group. The apelin expression of brain.heart,vascular and kidney tissues in hypertension rat increased significantly in the process of the hypertension in immunochemistry and ELASA. The content of apelin in brain were respectively 3.05 ± 0.902,5.06 ± 1.003,5.78 ±0.862,6.44±1.236,10.03±1.25(μg/mg protein) at 0, 2, 4, 8, 12w. The content of apelin in heart were 4.96±0.426,4.88±0.358,5.86±0.406,6.34±0.339 ,7.25±0.386 (10-1 μg/mg protein) at 0, 2, 4, 8, 12w respectively. The content of apelin in blood vessel were respectively 3.32± 0.186,3.66 ±0.247,4.36 ±0.235.5.49 ±0.268,6.04 ± 0.325(10-1 μg/mg protein) at 0, 2, 4, 8, 12w. The content of apelin in kidney were 3.32 ± 0.255,3.41 ± 0.297,4.36 ± 0.392.5.49 ± 0.33.5.58 ± 0.281(10-1 μg/mg protein) at 0, 2. 4, 8, 12w respectively. The ACE2 and caveolin-1 expression of...
