Background Cervical cancer is one of the most common malignancies,and over 90% cervical cancers are pathogenically associated with high risk HPV infection.In recent years,the morbidity and mortality of cervical cancers have kept increasing,and its treatment should be still more important.Capsaicin is one of the ingredients from red pepper.It's reported that its medical effects consist of pain relieving and anti-inflammation etc.Now many studies showed that capaicin can inhibit cancer growth and speed up cancer cell apoptosis,while the related mechanisms are still unclear.Objective To approach the inhibiting effects of capsaicin on cervical cancer cell and the mechanism of inhibiting effects,by the observation of the inhibition and apoptosis of 2 cervical cancer celllines under treatment of capsaicin,and by the detection of intracellular reactive oxygen species and expression of capsaicin receptor TRPV1 and related apoptosis genes.Methods Two kinds of cervical cancer cell lines,HPV positive Hela and HPV negative C-33 A cells,were chosen as studying subbjects.By cell culture and flow cytometry,the inhibition of cell growth and the affects of ROS and cell cycles and cell apoptosis in the 2 kinds of cancer cells,after capsaicin treatment,were examined.By means of q PCR and Wester Blot,we detected,in Hela and C-33 A cells after capsaicin treatment,the expression of capaicin recetor TRPV1,and the expression of cytochrom C,Caspase3,Bax,Bcl-2,P53 and P21 in the pathway of cell apoptosis.Results In Hela and C-33 A cells after capsaicin treatment,their inhibition of growth was getting severer along with the capaicin concentration increase and time prolongation,and the cell cycle ceased at stage G2/M.The capsaicin concentration of 200?M could significantly increase the ROS level in Hela cells(p<0.05),and the capsaicin concentration of 600?M could only significantly increase the ROS level in C-33 A cells(p<0.05).The capsaicin could promote the apoptosis of both kinds of cancer cells,and even more apoptosis of Hela cells.The expressions of capaicin recetor TRPV1 and the expressions of P53 and P21 were not significantly changed both at m RNA and protein level.The expression of cytochrom C and Caspase3 and the expression of bax were significantly increased,and the expression of Bcl-2 was significantly decreased.Theresults revealed that capsaicin do have the effects of repressing cell proliferation,and induce cell apoptosis by the increase of reactive oxygen species(ROS)in Hela and C-33 A cell.Conclusions Capsaicin could effectively inhibit the growth and promote the apoptosis of Hela and C-33 A.The anti-tumor effects ofcapsaicin might be achieved not by TRPV1 or by P53 pathway,but probably by Cytchrome C pathway.Specifically,capsaicin may induce ROS increase in Hela and C-33 A cell,then Cytchrome C relived from mitochondria and trigered Caspase 3 appoptosis pathway in cell.
