Objective Burkitt's lymphoma(BL)is a highly invasive B-cell lymphoma.Although there are now many treatments available to improve the prognosis of this rapidly growing lymphoma,treatments such as high-intensity chemotherapy still have significant treatment-related toxicity.Therefore,there is still a need to find new and effective treatment strategies.Uncontrolled regulation of mi RNAs is associated with many human diseases including lymphoma.Many human mi RNAs are located in tumor-associated genomic regions,and dysregulated mi RNAs may be involved in tumor formation as oncogenes or tumor suppressors.This preliminary study aimed to investigate the biological role that mi RNAs may play in the pathophysiology of BL.Methods In this study,mi RNAs that affect the proliferation of BL cells were screened by CCK8 method.Then,the expression level of the selected mi RNA in BL cells was further detected by Realtime PCR.The effect of mi R-150-3p on apoptosis and cell cycle of BL cells was detected by flow cytometry.The biological effects of mi R-150-3p in BL were further explored by colony formation assay.Finally,the biological effects of mi R-150-3p in different cells were examined,and its downstream targets were sought to explore the mechanism of its biological effects.Results The expression level of mi R-150-3p in Raji cells was reduced,and it could inhibit Raji cell proliferation and cell colony forming ability,and regulate the cell cycle of Raji cells.But mi R-150-3p does not affect the apoptosis of Raji cells.The expression of mi R-150-3p was decreased in CA46,but did not affect the proliferation,apoptosis and colony forming ability of CA46 cells.The expression of mi R-150-3p is also decreased in SNK6 cells,and it can reduce the proliferation of SNK6 cells and promote apoptosis.The ARPP21 and CDH2 genes have no significant direct interaction with mi R-150-3p,but it may be associated with mi R-150-3p related singnal transduction pathway.Conclusion mi R-150-3p produces different biological effects in EBV-positive BL cells,EBV-negative BL cells,and EBV-positive NKTL cells.This also suggests that the biological effects of mi R-150-3p may be related not only to the infection status of EBV,but also to the differentiation and development of B cells.
