Objective:Studying preservation effects and its mechanism of a new organ preservation solution-HPG solution in vitro.Methods:Forty beagle canines were divided equally into HPG group and LPD group,perfused with HPG solution and LPD solution respectively.After perfusion,the lung were preserved for 6-hour,12-hour and 18-hour at 4 ?.The tissue wet-to-dry ratio,apoptosis index,MPO,MDA and SOD were used as test indicators.And the changement of the lung structure under light microscope.Meanwhile,completing the left lung transplantation animal model.Blood gas analysis(preoperation,postoperation2 hours,postoperation 6 hours and postoperation 12 hours),chest X-ray at 8-hour after transplanting and the operating time were estimated the model.Results:Results 1:The tissues of HPG group and LPD group were damaged when they were preserved for a long time in vitro.About 6 hours late,both HPG group and LPD group,having a clear structure under light microscope,no exudation in the alveoli,no thickening of the alveolar wall,and no edema of the pulmonary interstitial.But,in conservation 12 hours and 18 hours,the degree of injury of the HPG group is lighter than the LPD group(P<0.05).The W/D ratio is not significant difference in HPG group and LPD group at 6-hour,but at 12 hours and 18 hours,HPG group is less than LPD group(P<0.05).The apoptotic index in HPG group is lower than the LPD group at any time(6-hour,12-hour and 18-hour)(P<0.05).whether the HPG group or the LPD group,there is been inflammatory damaged tissue in vitro.The HPG group is always less than the LPD group in MPO and MDA at 6-hour,12-hour and 18-hour,it makessense(P<0.05).Shorter storage,for example storage for 6 hours,the SOD has little difference in the group of two,but longer preservation,for example preserving 12 hours or 18 hours,the activity of HPG's SOD is stronger than the LPD's(P<0.05).Results 2: All beagle dogs had no anesthetic accident.The receptor canine ware survival after transplantation.The total operating time was(138±13)min;the recipient took(87±20)min,and(42±7)min was the cold ischemia time.The preoperation oxygenation index was(401.3±31.5),after 2 hours transplantation,the oxygenation index was(389.5±34.6),but the arterial oxygenation index became(339.4±34.7)at 6h postoperatively.The 6-hour decline rate is high,but it was still more than 300.And it maintains this downward trend to 12 h after surgery(oxygenation index 262.4 ± 27.3).The volume of transplanted lung espanded well at 8h after operation.Conclusion:Conclusion 1:The effects of preservation at 6 hours both HPG solution and LPD solution was same,but at 12 hours and 18 hours,the effect of HPG group was better than that of LPD group.It may be through enhancing clearing oxygen free radical,inhibiting neutrophil aggregation and activation,reducing cell apoptosis to achieve preservation purposes.Conclusion 2: The animal model of left lung transplantation in Beagle dogs was successfully established.This animal model is easy to replicate and has good feasibility.The animal model can provide a good platform for the research of lung preservation,pulmonary perfusion,and ischemia-reperfusion injury and so on.
