Objectives Detect the levels of autophagy related proteins Beclin 1,LC3 and P62 in missed abortion and artificial abortion villi tissues,and compare the differences between the two groups,so as to explore the relationship between autophagy activity and missed abortion.Methods From July 2018 to October 2018,20 cases of missed abortion patients who admitted to Hebei Provincial People’s Hospital were collected as the missed abortion group,and 20 cases of normal early pregnancy patients who required induced abortion due to unplanned pregnancy during the same period were collected as the normal pregnancy group.The villi were collected from the two groups of patients within a few minutes after hysterectomy or induced abortion respectively,and then washed with normal saline and stored under appropriate conditions.Immunohistochemistry and Western Blot were used to detect the expressions of autophagy related proteins Beclin 1,LC3 and P62 in the villi of the two groups,and the differences in the expressions of various proteins between the two groups were analyzed and compared.Results 1 Comparison of general conditions: there was no significant difference in the mean age and gestational age between the missed abortion group and the normal pregnancy group(P>0.05).2 Immunohistochemical detection: Beclin 1,LC3 and P62 were positively expressed in villi of missed abortion group and normal pregnancy group,and the brown particles were mainly located in the cytoplasm of villous trophoblast cells.Beclin 1 and LC3 in villi of missed abortion group were stronger than that of normal pregnancy group,while P62 was weaker than that of normal pregnancy group.The MOD value of Beclin 1 and LC3 in villi of missed abortion group was significantly higher than that of normal pregnancy group,and the difference between the two groups was statistically significant(P<0.05).The MOD value of P62 in villi of missed abortion group was lower than that of normal pregnancy group,the difference between the two groups was not statistically significant(P>0.05).3 Western blot assay: the expression levels of Beclin 1 and LC3-II/I were significantly higher than that of the normal pregnancy group,and the difference between the two groups was statistically significant(P<0.001).The expression level of P62 in villi of the missed abortion group was significantly lower than that of the normal pregnancy group,and the difference between the two groups was statistically significant(P<0.001).4 Correlation analysis of Beclin 1,LC3-II/I and P62 protein with age and gestational age: 1)Beclin 1,LC3-II/I and P62 were not significantly correlated with age(P>0.05).2)There was no significant correlation between gestational age and the expression level of Beclin 1 and P62 in villi(P>0.05).There was a negative correlation between the gestational age and the expression level of LC3-II/I in the villous tissue,which was statistically significant(P<0.05).Conclusions 1 Beclin 1,LC3 and P62 were mainly expressed in the cytoplasm of villus trophoblast cells.2 The expression of autophagy activity in missed abortion women was stronger than that in normal pregnant women,suggesting that the up-regulation of autophagy activity was associated with the occurrence and development of missed abortion.Figure 5;Table 4;Reference 139...
