Effect Of Zusanli Acupuncture On The Expression Of Autophagy-Related Protein(Atg) LC3 In Interstitial Cell Of Cajal (ICC) In Small Intestine Of Rats With Syndrome Of Functional Dyspepsia(FD)

Objective: Taking Interstitial Cell of Cajal(ICC)in Small Intestine of FD Rats as the object of study,this research aims to explore the effect of Zusanli acupuncture on the expression of autophagy-related protein(Atg)of LC3 in ICC via quantitative analysis on the expression of autophagy-related protein(Atg)of LC3 in ICC.Methods: 25 male and 25 female adult Wistar rats of SPF(specific pathogen free,SPF)grade were selected and randomly divided into 5 groups including normal control group(Group A,n=10),FD model group(Group B,n=10),natural recovery group(Group C,n=10),false acupuncture group(Group D,n=10),acupuncture at Zusanli point group(Group E,n=10).In group A,natural feeding with nothing special treatment was given.In the group of B,C,D and E,Functional Dyspepsia(FD)model was applied to all rats,which were established in the way of Chinese cool and relieving stagnant Qi medicine gavage.Thereafter,Group D and E received sham electro acupuncture treatment and acupuncture at Zusanli point treatment separately.Observing and recording changes in general biological behavior and weight of rats in each group in the process of modeling and treatment.After modelling and treatment,test urinary D-xylose excretion rate and intestinal propulsive rate in each group.Their small intestine adjacent to the pylorus were eviscerated for experimental study.Quantitative analysis on expression of LC3 via western blotting was conducted to explore the the effect of Zusanli acupuncture on the expression of autophagy-related protein(Atg)of LC3 in ICC.Results:1.Compared with control group A,a significant increase in the ratio of LC3-II to LC3-I was found in model groups including group B,C,D and E in varying extent(P<0.05=;2.In model groups including group B,C,D and E,an obvious decrease in the ratio of LC3-II to LC3-I was detected in acupuncture at Zusanlipoint group E and the statistical difference was significant(P<0.05=;3.There was no significant difference in the ratio of LC3-II to LC3-I among Group B,C and D(P>0.05).In general,the basic trend in expression quantity of Atg8/LC3 was: FD model group B ≈natural recovery group C ≈ sham acupuncture group D > acupuncture at Zusanli point group E > control group A.Conclusion:1.Zusanli acupuncture can decrease the expression of autophagy-related protein(Atg)of LC3 in ICC of FD rats;2.Zusanli acupuncture may improve symptoms of Functional Dyspepsia(FD)through regulating the level of Interstitial Cell of Cajal(ICC)autophagy.
