Objective: To investigate the expression of CYBA gene coding p22 phox subunit of nicotinamide adenine dinucleotide phosphate oxidase in pre-eclamptic placenta and the relationship between the C242 T single nucleotide polymorphism of the CYBA gene and severe pre-eclampsia in Shandong province,to provide a new theoretical basis for the pathogenesis of pre-eclampsia.Methods: Firstly,the placenta of pre-eclampsia and healthy pregnant women who were hospitalized and delivered in the obstetrics department of the Affiliated Hospital of Qingdao University from June 2017 to October 2018 were collected.21 cases of pre-eclamptic placenta and 21 cases of healthy placenta were randomly selected as subjects.Total RNA and protein were extracted from placenta tissue,and the expression of CYBA gene was detected by relative fluorescence quantitative PCR and Western-Blot method.Data analysis was performed using Graphpad Prism software.Then,the peripheral blood of pregnant women who were hospitalized in the obstetrics department of the Affiliated Hospital of Qingdao University,Shandong Provincial Hospital,and the Affiliated Hospital of Binzhou Medical College from January 2014 to December 2017 were collected.Using the logit model of R language(PS±0.2SD),the nationality,age,body mass index(BMI index),pregnancy time were matched with the propensity score of 1:1.2 samples,and 1184 cases of severe pre-eclampsia and 1421 cases of healthy control were selected.The DNA of peripheral venous blood of above 2605 subjects was extracted,and the C242 T polymorphism of CYBA gene was genotyped by TaqMan probe method.P < 0.05 indicates that the difference was statistically significant.Results: 1.Compared with the control group,the expression of CYBA mRNA and protein in severe pre-eclamptic placenta was significantly increased and the difference was statistically significant(P<0.05).2.There was no significant difference in the frequency distribution of CYBA gene C242 T genotype and allele frequency between pregnant women with severe pre-eclampsia and healthy controls(P>0.05).There were no significant differences in genotype and allele frequency distribution between early-onset severe pre-eclampsia pregnant women,late-onset severe pre-eclampsia pregnant women and healthy control pregnant women(P>0.05).Conclusions: 1.The CYBA gene is highly expressed in the placenta of pre-eclampsia,indicating that it may be involved in the development of pre-eclampsia.2.There was no correlation between C242 T polymorphism and severe pre-eclampsia in Shandong province,but this result may be related to the population,region and selection criteria of the study population.Therefore,in the future,a larger sample size will be needed for verification,and a new genetic susceptibility gene study will be actively conducted to explore the pathogenesis of pre-eclampsia.
