Objective:Testingexpression level of miR-4534 in the serum of patients with non-small-cell lung cancer(NSCLC),it is to aim to explore whether the miR-4534 can be used as a non-small cell lung cancer invasion and metastasis ability evaluation index,and to explore the clinical relationship between abnormal expression of miR-4534 and the common influence clinical features of invasion and metastasis in NSCLC patients and to analysis whether the differences of miR-4534 expression level has the influence in the prognosis of patients with non-small cell lung cancer at the same time?Methods:We screen miRNAs with differential expression in serum of patients non-small cell lung cancer patients and normal serum by gene chip technology at high throughput.We choosethe significantly over expression levelof miR-4534 in non-small cell lung cancer serum to study.At present,real-time fluorescence quantitative PCR(qRt-PCR)was applied to verify the expression level of miR-4534 in serum of patients with non-small cell lung cancer(NSCLC).Serum samples from 40patients with NSCLC and 40 normal subjects were enrolled according to certain conditions.SPSS 22.0 statistical analysis software was used for correlation statistical analysis.Analysising the expression of miR-4534levelsBy 2~-??Ct??Ct method.Independent sample T test was used for the difference between groups.Using ROC curve analysis miR-4534 used as the evaluation indexofsensitivity and specificity in non-small cell lung cancer invasion ability.The overall survival in Non-small cell lung cancer is Analysised between miR-4534 high expression group and low expression group with Kaplan Meier method.It has statistical significance.at P value?0.05.Results:1?The expression level of miR-4534 in serum was significantly different between the non-small cell lung cancer group and the normal group(P<0.05),and the expression level of miR-4534 was statistically up-regulated in non-small cell lung cancer(P<0.05).2?The expression level of miR-4534 in efficiency of the distinguish between normal person and patients with NSCLC,the area under the curve is 0.928,and the sensitivity is 92.5%,and the specific degree is85%.The expression level of miR-4534 in efficiency of distinguish between normal personand in?/?stage NSCLC patients,the area under the curve is 0.82,and the sensitivity is 77.5%,and the specific degree is85.7%.The expression level of miR-4534 in efficiency ofdistinguish?/?period and normalperson,?/?NSCLC patients,the area under the curve is 0.97,and the sensitivity is 87%,and the specific degree is 100%.3?MiR-4534 was significantly correlated with smoking,Lymph node metastas,distant metastasis,cancer size and clinical stage in the serum of non-small cell lung cancer(P<0.05),and had no correlation with pathological classification,age or gender(P>0.05).4?The median survival time of the miR-4534 high-expression group in the serum of patients with non-small cell lung cancer was 21 months,and that of the low-expression group was 26 months,respectively.Therefore,the survival time of serum miR-4534 in patients with non-small cell lung cancer was shortened compared with that in the low expression group(P<0.05).Conclusion:MiR-4534 can be used as a judgment of non-small cell lung cancer invasion and metastasis of markers.It was significantly correlated with smoking Lymph,node metastas,distant metastasis,cancer size and clinical stage in the serum of non-small cell lung cancer.It was used as judging the prognosis of patients with non-small cell lung cancer.
