Abstract/Summary:
Objective Through in vitro experiments,the effects of different concentrations of propofol on the activation ability of peripheral nerve schwann cells were observed from three aspects of proliferation,migration and invasion,and to explore the potential mechanism of propofol promoting the repair of the peripheral nerve injury.Methods The frozen Schwann cells were thawed,sub-cultured,and taken into treatment during the logarithmic growth phase.The cells were digested by trypsin and re-suspended in the complete medium,the cells were counted before treatment.The cells were divided into control group(C)and experimental group(P),Group C schwann cells cultured with completely without propofol medium,P group of schwann cells with different concentrations of propofol 20 μM(P2),40 μM(P3),80 μM(P4)set of nutrient solution culture.1.The proliferative activity of Schwann cells was detected by CCK-8 method: The cells inoculated on 96-well culture plate,each group had three pores,which were set for 12 hours,24 hours and 48 hours respectively,The absorbance of Schwann cells cultured with different concentrations of propofol was measured by CCK-8 method.2.Scratch test was used to detect the migration of Schwann cells: Cells were counted and inoculated on 6-well plate,Scratches were made with a liquid transfer gun after the cells were covered with the hole plate,photographs were taken at two time points of 0 hours and 24 hours respectively,observe the distance between the scratches.3.The invasion ability of Schwann cells was detected by transwell chamber: After counting,the cells were inoculated in the upper chamber of transwell,and the lower chamber was placed in the DMEM medium containing 20% serum.After 24 hours of culture,the invasive cells were counted.4.Results 1.Compared with the control group,the proliferative capacity of Schwann cells in P2,P3 and P4 groups were increased(P<0.05),of which P2 and P4 groups were significantly lower thanthe P3 group(P<0.05).2.Compared with the control group,the Schwann cells in P2,P3 and P4 groups showed increased migration capacity(P<0.05),of which the P2 and P4 groups were significantly lower than the P3 group(P<0.05).3.Compared with the control group,the number of invasive Schwann cells in P2,P3 and P4 groups increased(P<0.05),of which the P2 and P4 groups were significantly lower than the P3 group(P<0.05).Conclusions 1.Different concentrations of propofol is able to promote the proliferation,migration and invasion of Schwann cells,and the most effective concentration of propofol is 40 μM.2.Propofol may promote the repair of peripheral nerves injury by promoting the activation of Schwann cells.
