[Objective]Colorectal cancer(CRC)is a common global malignancy.Relevant statistics show that the cases of clinically diagnosed colorectal cancer in China are getting younger and younger.This situation has a certain relationship with the rapid development of domestic economy in the past decade,and its incidence rate is 1/5 of the total number of people under the age of 40,and it has a tendency to continue to rise year by year.In the world,China has entered the ranks of high incidence of colorectal cancer.In addition,colorectal cancer is difficult to detect and treat in time,which often leads to the middle and late stage after the onset of the disease,which causes many difficulties for the prognosis and treatment of patients in the future.The increase in the incidence of colorectal cancer and the high mortality caused by the increased difficulty of its discovery seriously threaten the health of our people.With the advancement of science and technology,the development of clinical treatment combined with the further maturity of cancer-targeted therapy,this has improved the treatment difficulty and prognosis of patients with colorectal cancer to some extent.However,in patients with advanced colorectal cancer and metastasis,the prognosis remains poor.In order to achieve the goal of increasing the survival rate of patients with colorectal cancer,medical workers have been trying to explore the pathogenesis of CRC and its corresponding treatment strategies.mi RNAs(micro RNAs)are defined as a class of non-coding RNAs that are short in length(total length of the fragment is about 18-22 nt)and can regulate the expression of related genes after transcription.Studies have shown that there are multiple mi RNAs expression levels and changes in their multiple related genes in colorectal cancer cells.mi RNAs can directly target a variety of oncogenes,thereby exerting their effects on the biological functions of cancer cells,such as the cell activity,migration,invasion,and proliferation of cancer cells.However,its specific complex regulatory mechanisms are not fully understood.We hypothesize that mi RNAs affect various biological functions of colorectal cancer cells because they can target and regulate related oncogenes.The aim of this study was to investigate the effect of mi R-621 and its regulation of its corresponding target genes on the development of colorectal cancer cells.[Methods] The mi R-621 was selected as the research object.The expression of mi R-621 in colorectal cancer cells was verified by q RT-PCR.The plasmid pc DNA3/pri-mi R-621 was constructed and verified by MTT,transwell migration and colony experiments.Inhibition of cell function in colorectal cancer cells.After mi R-621 related target gene was predicted by bioinformatics,and the EGFP plasmid of target gene was constructed,Western blot analysis confirmed that mi R-621 can directly target the corresponding target gene RPL15,and then determine the mi R-621 by Western blot.The effect of its target gene expression.Then,the effects of target genes on the biological function and EMT process of colorectal cancer cells were verified by MTT,transwell migration assay and colony assay and Western blot.Finally,a rescue experiment was designed to validate the effects of mi R-621 and its target genes on the biological function of colorectal cancer cells.[Results] The sequencing results showed that miR-621 was expressed in colorectal cancer tissues lower than its corresponding paracancerous tissues;mi R-621 could inhibit the function of colorectal cancer cells;bioinformatics prediction showed that RPL15 is a direct target gene of mi R-621,After constructing its corresponding EGFP vector,Western blot analysis showed that mi R-621 can directly target RPL15,and mi R-621 can inhibit the expression of target gene RPL15 to some extent.Rectal cancer phenotype experiment proves that target gene RPL15 can promote colorectal Cellular activity,migration and invasion ability and proliferation ability of cancer cells;design rescue experiments have demonstrated that mi R-621 not only inhibits the biological function of colorectal cancer cells by itself,but also increases its inhibition by affecting the expression level of its target gene RPL15.ability.[Conclusions] mi R-621 can inhibit the biological function of colorectal cancer cells,such as inhibition of its cell activity,migration and invasion ability and proliferation ability.It can also target RPL15 and inhibit the expression of RPL15.RPL15 can promote the corresponding biological functions of colorectal cancer cells,so the tumor suppressive effect of mi R-621 is achieved to some extent by inhibiting the expression of its target gene RPL15.The experimental results in this study demonstrate that mi R-621 can inhibit the expression of colorectal cancer,and can also enhance the effect of CRC on the expression of the gene RPL15,inhibit the occurrence and development of colorectal cancer,and clarify its specific role and mechanisms.
