Objective: To investigate the expression of perforin and granule enzyme B in patients with brucellosis in Inner Mongolia Autonomous region,and to provide a powerful basis for the determination of cellular immune function and basic research in patients with brucellosis.Methods:1?Participants: all patients were diagnosed as brucellosis from December 2016 to December 2018.The patients were treated in the affiliated Hospital of Inner Mongolia Medical University,aged 52.40 ±8.64 years old.There were 80 patients(45 males and 35females)with the age of 52.40 ±8.64 years old.A total of 60 cases(26 males and 34 females),aged 51.33 ±8.75 years,were selected from the affiliated Hospital of Inner Mongolia Medical University in the same period.No symptoms,signs and plate agglutination tests were negative for brucellosis.There was no statistical difference in age and sex between the two groups(P>0.05).2.Methods : In this study,the expression of perforin and granzyme B mRNA in lymphocytes of patients with brucella was detected by real-time fluorescence quantitative method,and the software SPSS for Windows20.0 was used.T test was used to describe the expression of perforin and granzyme B in the patients group and the control group,the age of the two groups was independent sample t test,? 2 test was used in the two groups,the difference was statistically significant(P < 0.05).Results: the NKG2 D,perforin and granzyme B genes were amplified by using designed primers and showed specificity by agarose gel electrophoresis and dissolution curve analysis.The results of GreenI real-time fluorescence quantitative PCR showed that the expression of perforin and granzyme B in patients with brucellosis was significantly higher than that in normal controls(P < 0 05).Conclusion :The expression of perforin and granzyme B in peripheral blood lymphocytes of patients with brucellosis increased,suggesting that the expression of perforin and granzyme B may play an important role in the immune response to Brucella infection in the pathogenesis of brucellosis.
