Objective The incidence of papillary thyroid cancer(PTC)has been rapidly rising around the world,some PTCs with invasion and metastasis are refractory with poor prognosis.But the pathogenic mechanism is unclear.Currently lncRNA has been found closely related to the development of occurrence and development of some tumors through the regulation of the target genes.This study aims at finding out the abnormal lncRNAs related to PTC with local invasion and metastasis.Investigating the biological processes of lncRNAs in order to further study on molecular mechanism of PTC with invasion and metastasis.Methods In our study,The use of lncRNA microarray gene chip firstly screened abnormal expression of lncRNA and mRNA genes between PTC tissues and paired adjacent noncancerous thyroid tissues.Then we study on the biological function of differentially expressed lncRNA and mRNA by bioinformatics analysis methods and construct the co-expression networks,in order to explore the role that lncRNA played in regulation of PTC.We further investigated the interaction of lncRNA-miRNA-mRNA through ceRNA network analysis.Results The expression profile of lncRNA was studied by lncRNA microarray gene chip,and 338 different lncRNAs genes were discovered in the PTC tissue with local invasion and metastasis compared with normal adjacent thyroid tissue.Bioinformatics analysis predicted the function of differentially expressed genes.The ceRNA networks were constructed,and our data showed the network nodes between 29 lncRNA and 67 coding genes.The real-time PCR showed a up-regulation of EphA4(p<0.05)and lncRNA RP11-399O19.8 compared with normal tissue(p<0.05),while let-7a-5p down-regulated in tumors.(p<0.05).Conclusion Our study reveals a complex relationship of lncRNAs and mRNAs,and lncRNAs could potentially mediate protein-coding genes expression as ceRNA to contribute to the development of PTC with invasion or metastasis.Clarification of the mechanism is beneficial to explore new therapeutic targets for refractory PTC.
