| Ionizing radiation consists of artificial radiation and natural radiation.Ionizing radiation could cause a series of reactive oxygen species(ROS)in the body,then leads to a series of biological macromolecules such as proteins,nucleic acids and lipids damage.Water is ionized to free radicals,and constantly attack the body,especially immune organ that protects the body from external damage,eventually lead to a vicious cycle.Radiation could cause oxidative stress,thus the protective effect is related to antioxidant and immune improving.Studies have shown that Auricularia auricula polysaccharide and Pinus koraiensis polyphenols have strong antioxidant activity and immune improving activity.This subject screened the polysaccharide and polyphenols from Auricularia auricula and Pinus koraiensis through the antioxidant activity in vitro,the AAP-3 and PKP-2 was screened out and structural identified;the synergy effect of AAP-3 and PKP-2 was evaluated through Chou-Talalay theory;systematically studied the in vitro protective effect of AP on radiation induced oxidative stress.Twelve segments of Auricularia auricula polysaccharides were isolated with different pH value solvent and graded by different concentration of ethanol,four components of Pinus koraiensis polyphenols were elution by D101.The scavenging activity on hydroxyl radicals and ABTS radical,reducing power,lipid peroxidation inhibition activities of segments and components were studied.The results indicated that AAP-3 and PKP-2 had the most significant antioxidant activities.The AAP-3 was purified by DEAE-52 and characterized by UV,FT-IR,GC-MS.The results suggested that AAP-3 was composed of ribose,arabinose,xylose,mannose,glucose and galactose with a ratio of 0.1:1:1:3:0.7:0.1.The contents of PKP-2 were measured by determination of polyphenols,flavonoids,procyanidins,FT-IR and HPLC.The results indicated that taxifolin was the main composition.According to the LD50,AAP-3 was combined with PKP-2 with a ratio of 3:1,the antioxidant activity of AAP-3 and PKP-2 was evaluated through Chou-Talalay theory.The results indicated that the Combination Indexes(CI)were less than 1,the synergy effect of AAP-3 and PKP-2 on scavenging activity on hydroxyl radicals and ABTS radical,reducing power,lipid peroxidation inhibition activities was certain.The Dose-Reduction Indexes(DRI)were more than 1,indicated that AAP-3 in combination with PKP-2(AP)reduced the dose.The oxidative damage model of mice induced by 60CO-γ radiation was established to investigate the radioprotective effect of AP.The results showed that AP could effectively increase normal white blood cell counts,liver index,heart index,spleen index,splenocytes proliferation ability,phagocytic index;reduce brain index.There were no significant difference with Normal group(P>0.05)and significant difference with Model group(P<0.05),indicated that AP could protect the immune system against damage induced by radiation.The results showed that AP could effectively reduce the level of MDA in serum and organs;increase the activity of SOD,GSH-Px,CAT and the content of Vc,VE,GSH,CoQ10 in serum and organs.There were no significant difference with Normal group(P>0.05)and significant difference with Model group(P<0.05),indicated that AP could protect antioxidative system against damage induced by radiation.The results showed that AP could effectively reduce bone marrow micronucleus rate,rate of chromosome aberration of bone marrow;increase DNA content of bone marrow.There were no significant difference with Normal group(P> 0.05)and significant difference with Model group(P< 0.05),indicated that AP could protect DNA against damage induced by radiation.The oxidative damage model of mice induced by 60Co-γ radiation was radiation.The results showed that AP could effectively increase normal white blood cell counts,liver index,heart index,spleen index,splenocytes proliferation ability,phagocytic index.It indicated that AP could protect the immune system against offspring damage induced by radiation.The results showed that AP could effectively reduce the level of MDA in serum and organs;increase the activity of SOD,GSH-Px,CAT in some degree and the content of VC,VE,GSH,CoQ 10 in serum and organs.It indicated that AP could protect antioxidative system against offspring damage induced by radiation.The results showed that AP could effectively reduce bone marrow micronucleus rate,rate of chromosome aberration of bone marrow,comet tail lengths,the number of cells in G0/G1 phase;increase DNA content of bone marrow.It indicated that AP could protect DNA against offspring damage induced by radiation.These results indicated that AP could combine the great antioxidant activities of AAP-3 and PKP-2,show synergy effect on protecting immune system,antioxidative system,DNA against damage induced by radiation both in mice and offsprings.The present study will play an important and practical role in revealing protective mechanism of radiation and exploiting the natural and efficient raidioprotectors. |
