Objective: To further elucidate the therapeutic effect and mechanism of MEBT/MEBO on chronic refractory wound by investigate the expression level of protein CK10,CK14 and CK10,CK14 mRNA in chronic refractory surface wound of rats.Methods: In this study,90 SPF Wistar rats were randomly divided into five groups: blank group(18 rats),acute injury group(18 rats),Chronic wound group(18 rats),rb-bFGF group(18 rats)and MEBO group(18 rats).The wound was treated according to the requirements of each group and the wound tissue was taken on the 3rd day,7 day and 14 th day after modeling.After all sample are collected,on the one hand,to compared the morphological differences of the wounds of rats in each group by Masson stain;on the other hand,using Western Blotting,RT-PCR method to detect the protein and gene expression of CK10 and CK14.Results:(1)There was no obvious change in the morphology of the wound tissue of rats in the blank group under the microscope.With the prolongation of treatment cycle,the edema of wound tissue in rats gradually subsided,the number of new blood vessels increased significantly,and the appendages of skin gradually formed.MEBO group and bFGF group was significantly better than the Chronic wound group In the healing process.(2)Western Bloting: On day 3,7,14 after local treatment,no significant change in expression level of protein CK10,CK14 in back skin tissues of rats in blank group was found and the difference were statistically not significant(P>0.05).Significant changes in expression level of protein CK10,CK14 in back skin tissues of rats in acute injury group,Chronic wound group,rb-bFGF group and MEBO group were found and the differences were statistically significant(P<0.05).the expression level of protein CK10,CK14 in wound tissues of rats showed a trend of increase.On day 7,14 after local treatment,expression level of protein CK10,CK14 in rats' wound of rb-bFGF group and MEBO group was higher than Chronic wound group and the comparison showed statistically significant difference(P<0.05).(3)RT-PCR: On day 3,7,14 after local treatment,no significant change in expression level of CK10,CK14 mRNA in back skin tissues of rats in blank group was found and the difference were statistically not significant(P>0.05).Significant changes in expression level of CK10,CK14 mRNA in back skin tissues of rats in acute injury group,Chronic wound group,rb-bFGF group and MEBO group were found and the differences were statistically significant(P<0.05).the expression level of CK10,CK14 mRNA in wound tissues of rats showed a trend of increase.On day 7,14 after local treatment,expression level of CK10,CK14 mRNA in rats' wound of rb-bFGF group and MEBO group was higher than Chronic wound group and the comparison showed statistically significant difference(P<0.05).Conclusion:(1)MEBO can promote the healing of chronic wound in rats.(2)MEBO can increase the expression levels of protein CK10,CK14 and CK10,CK14 mRNA in wound tissues.(3)MEBT/MEBO can promote epidermal stem cells proliferation and differentiation into normal epidermal cells,and then play a certain role in the repair of chronic refractory wounds.And its effect is no less than rb-bFGF group.
