The Role Of NCX1 In The Development Of Non-small Cell Lung Cancer

Objective: This study designed to explore the mechanism of NCX1 in the development of non-small cell lung cancer.Methods:1 Cell experiment:Using RT-qPCR and Western Blot to detect the expression of NCX1 mRNA and NCX1 in the cell of A549 and BEAS-2B.2 Tissue experiment:1)28 cases of non-small cell lung cancer tissues and 28 cases of its tumor adjacent histologically normal tissues were collected.2)Using RT-qPCR and Western Blot to detect the expression of NCX1 mRNA and NCX1 in the tumor tissue and normal tissue,analyzing the relationship between the expression of NCX1 and patients' gender,age,smoking history,and tumor metastasis.3)The expression of NCX1 protein were determined by immunohistochemistry.3 Statistical methods used in this study: SPSS 24.0 software package was used for statistical analysis:1)using t-test: measure data and conform to normal distribution and variance.2)using Mann-Whitney-Wilcoxon Test: measure data does not conform to the normal distribution.3)using the two-side test method,the difference being statistically significant at P<0.05.Results:1 Cell experiment: the expression of NCX1 mRNA and protein in A549 are higher than in BEAS-2B(P<0.05).2 Tissue experiment:1)RT-qPCR results showed that NCX1 mRNA expression was observed in both tumor tissues and normal tissues,but the mRNA and protein expression level of NCX1 in the tumor tissues of the subjects was significantly higher than that in the tumor adjacent histologically normal tissues adjacent to tumor(P<0.05).The expression level of NCX1 mRNA was correlated with the gender,smoking history,distant metastasis and type of non-small cell lung cancer: the expression of NCX1 mRNA in male NSCLC patients was higher than female patients(P<0.05).The expression of NCX1 mRNA in smoking patients was higher than that in non-smoking patients(P<0.05).In patients with distant metastasis,the expression of NCX1 mRNA was higher than that in patients without distant metastasis(P<0.05).The relative expression level of squamous cell carcinoma was higher than that of adenocarcinoma.There was no correlation between NCX1 mRNA expression level and the age(P>0.05)of NSCLC patients.2)Western Blot showed that NCX1 expression in NSCLC tissues was higher than that in corresponding tumor adjacent histologically normal tissues.3)Immunohistochemical results showed that NCX1 protein expression was observed in both non-small cell lung cancer tumor tissues and tumor adjacent histologically normal tissues,but NCX1 protein expression in non-small cell lung cancer tissues was generally higher than its corresponding normal control.Conclusions:1 NCX1 may be closely related to the occurrence and development of NSCLC.2 Tobacco may induce NCX1 expression.3 NCX1 has a certain relationship between NCX1 and the clinical prognosis of NSCLC.
