Objective To observe the effect of Astragalus Ramulus Cinnamomi Granule on the expression of mitogen-activated protein kinase(MAPKs)in myocardium of SD diabetic rats,and to explore the effect of Astragalus Ramulus Cinnamomi Granule on diabetic myocardium.Method Male SD rats were fed with high fat and sugar and intraperitoneally injected with streptozotocin to establish the model of type 2 diabetes mellitus.The rats were randomly divided into model group,positive control group,low dose group of Astragalus Ramulus Cinnamomi Granule,medium dose group of Astragalus Ramulus Cinnamomi Granule and high dose group of Astragalus Ramulus Cinnamomi Granule.After successful modeling,Guizhi and Guihuangqi Granules were given corresponding doses in low,medium and high dose groups,metformin hydrochloride was given in control group,and distilled water was given in blank group and model group.The body weight and fasting blood sugar of rats were recorded.At the end of the drug intervention period,rats were sacrificed and cardiac mass index was calculated;myocardial tissue sections were stained with HE to observe the pathological changes of myocardial tissue in each group;blood samples were taken from abdominal aorta to detect various biochemical indicators;and the expression of MAPKs-related protein was detected by immunoblotting.Results 1.Compared with the blank group,the expression level of MAPKs protein in the model group was significantly increased(p < 0.05);compared with the model group,the expression levels of histone in the control group and each dose were decreased in varying degrees(p < 0.05).2.Compared with the blank group,the HDL in the model group was significantly lower(P < 0.05),and the other biochemical indicators were increased in varying degrees.Compared with the model group,the HDL in the control group and the high dose group were significantly higher(P < 0.05);the other indicators were reduced in varying degrees.3.Compared with the model group,the weight of rats in the control group,the middle dose group and the high dose group decreased significantly(p < 0.05);compared with the model group,the fasting blood sugar in the high dose group and the control group decreased significantly(p < 0.05),but there was no significant difference between the low dose group and the control group(p > 0.05);compared with the normal group,the heart mass index in the model group increased significantly(P < 0.05),which was higher than that in the model group.There was significant difference between the dose group and the control group(P < 0.05).Conclusion 1? Astragalus Ramulus Cinnamomi Granule can affect the expression of MAPKs-related proteins.By inhibiting the expression of p-p38 MAPK,p-ERK and p-JNK,Astragalus Ramulus Cinnamomi Granule can inhibit the expression of p38 MAPK,ERK and JNK and alleviate the diabetic cardiomyopathy.2? Astragalus Ramulus Cinnamomi Granule can appropriately reduce TC,TG,LDL and increase HDL,at the same time reduce CK,LDH,HBDH,which may be one of the reasons for Astragalus Ramulus Cinnamomi Granule to alleviate myocardial lesions in rats.3? Astragalus Ramulus Cinnamomi Granule has the effect of controlling body weight and blood sugar in rats,but the effect of high dose and low dose is quite different,which indicates that the effect of Astragalus Ramulus Cinnamomi Granule requires higher dose.
