OBJECTIVE: To evaluate the expression of long noncoding RNA UCA1 in cervical exfoliated cells of cervical cancer and CIN patients.PATIENTS AND METHODS: Total 99 cervical exfoliated cells and cervical tissues were obtained for this study.Patients were divided into 3 different groups,cervical cancer,CIN and normal control groups with each group consisting of 33 cases respectively.UCA1 expressions in cervical exfoliated cells were measured by quantitative reverse transcriptionpolymerase chain reaction(q RT-PCR).The data were analyzed by SPSS 20.The UCA1 expression levels in cervical cells of three groups were compared.The associations of UCA1 in different clinicopathological features of cervical cancer cells were assessed.To determine the diagnostic value of lnc RNA UCA1 in cervical cells the receiver operating characteristic(ROC)analysis was evaluated in cervical cancer vs normal groups and CIN vs normal groups.The expression of UCA1 in cervical cells were compared with the immunohistobiomarkers p16INK4 A,Ki-67,HPV E6/E7 mRNA expression and UCA1 expression in cervical tissue to investigate the clinical values of UCA1 in cervical cells to monitor cervical cancer and CIN.RESULTS: UCA1 level is significantly high in cervical cancer cells compared to CIN cells(P<0.05)and normal cells(P<0.001).The expression level of UCA1 in cells of CIN was also higher than that in normal group(P<0.01).UCA1 in cervical cells could accurately discriminate cervical cancer from CIN and normal subjects.Elevated expression of UCA1 in cervical cells was correlated with advanced FIGO stage,differentiation and lymph node metastasis(P<0.05).ROC curve showed UCA1 in cervical cells is a good candidate to discriminate the difference between cervical cancer and normal subjects,with area under curve(AUC)of 0.764(95% CI: 0.649-0.879)and as well as CIN from normal subjects,with AUC of 0.707(95% CI: 0.575-839).A significant positive correlation was observed between UCA1 expression in cervical cells and immunehistobiomarkers p16INK4 A,Ki-67,HPV E6/E7 mRNA expression and UCA1 expression in cervical tissue.CONCLUSIONS: The high expression of UCA1 in cervical exfoliated cells of cervical cancer and CIN has a certain correlation with the occurrence and development of the disease,which provides a theoretical basis for the further search for new diagnostic and therapeutic markers of cervical cancer.
