Objective:The aim of this study is to investigate the expression of the Long non-coding RNA urothelial carcinoma antigen 1 (Lnc RNA UCA1) in cervical cancer tissues and its biological function in cervical cancer cells.Methods:Expression of UCA1 was analyzed by quantitative realtime polymerase chain reaction technique. The difference of UCAl expression with different pathological types. FIGO stages, depth of tumor invasion were analyzed. Small interfering RNA (siRNA) was used to suppress the expression of UCA1 or HPV E6 in cervical cancer cells. In vitro, assays were performed to explore the biological functions of UCA1 in cervical cancer.Results:1. There was found that UCA1 expression was markedly upregulated in the cervical cancer tissues than in the corresponding adjacent tissues and the nonneoplastic epithelial tissues(P<0.05). The expression was also upregulated in the corresponding adjacent tissues than in the nonneoplastic epithelial tissues(P<0.05). In addition there was found that UCA1 expression was upregulated in cervical intraepithelial neoplasias than in the nonneoplastic epithelial tissues(P<0.05).2.The expression of UCA1 in the cervical cancer tissues was significantly correlated with pathological types. FIGO stage and depth of tumor infiltration. The UCA1 expression of squamous cell carcinoma tissues was higher than adenocarcinoma tissues (P<0.01). The UCA1 expression of tissues from the patients in FIGO III was higher than the patients in FIGO II(P<0.01), and the expression of tissues from patients in FIGO II was higher than the patients in FIGO I(P<0.01). Besides if tumors had infiltrated all layers of cervix, there was higher UCA1 expression in the cervical cancer tissues (P=0.05).3.In vitro assays our data indicated that knockdown of UCA1 suppressed cell proliferation and changed cell cycle.4. In vitro assays, the knockdown of human papilloma virus 16 (HPV 16) E6 suppressed the expression of UCA1.Conclusion:Lnc RNA UCA1 expression is upregulated in the tissues of cervical cancer which is associated with pathological type. FIGO stage and depth of tumor infiltration. In addition, Lnc RNA UCA1 can promote cell proliferation,change cell cycle and it may be elevated by HPV E6.
