Background and Objective Lung cancer has become the most malignant tumor with the highest morbidity and mortality in the world in recent years.Estrogen plays an important role in promoting the progression of lung cancer.Tumor markers have a great significance in early diagnosis,pathological typing and tumor staging of lung cancer.However,there are few studies on the correlation between lung cancer tumor markers and sex hormone.This study was to investigate the significance of combined detection of estrogen(17?-Estrogen,E2)and tumor markers in the diagnosis and prognosis of lung cancer.Methods A total of 174 blood samples from females were collected from the Department of Thoracic and Cardiovascular Surgery of Zhongnan Hospital of Wuhan University from November 2012 to September 2014.Of these,117 patients had NSCLC and 57 patients had BPL,and postoperative pathology confirmed these results.An enzyme-linked immunosorbent assay was used to detect the expression of E2,carcinoembryonic antigen(CEA),neuron-specific enolase(NSE),and cytoker-atin 19 fragment antigen 21-1(CYFRA21-1)in patients with NSCLC and BPL to analyze the correlation between E2 and CEA,NSE,and CYFRA21-1 expression,and its correlation with clinicopathological features and prognosis.Normal lung epithelial cells BEAS-2B and lung cancer cells(A549,H1975,H460,SK-MES-1)were subcultured.Four lung cancer cells were treated as follows: blank control group,E2 group,Fulvestrant(Ful,anti-estrogen)group,E2+Ful group.The expression of tumor markers were detected by real-time reverse transcription-polymerase chain reaction(RT-qPCR).Western-blot was used to detect CEA,NSE,CYFRA21-1,and estrogen signaling pathway proteins such as mitogen-activated protein kinases(MAPK)and protein kinase B(AKT/PKB),p-p44/42 MAPK,p-AKT expression levels.The experimental results were statistically analyzed with SPSS22.0 software.The ?2 test was used to analyze the expression of E2 and tumor markers in NSCLC and BPL patients.The expression of E2 and tumor markers in different lung cancer cells was statistically analyzed using one?way analysis of variance.If p<0.05,the difference was considered statistically significant.Results The expression of E2,CEA,NSE and CYFRA21-1 in the serum of patients with NSCLC was significantly higher than that in patients with BPL(all p<0.05).The positive expression rate of E2,CEA,NSE and CYFRA21-1 in patients with NSCLC(73.50%,82.91%,83.76%,85.47%)was significantly higher than that in patients with BPL(26.32%,29.82%,33.33%,19.30%)(p=0.000).At the same time(Table 2),a positive correlation between E2 and the expression of CEA,NSE and CYFRA21-1(p=0.007,0.000,0.000)was observed.The higher the concentration of E2 in the serum of patients with NSCLC,the higher the concentrations of CEA,NSE and CYFRA21-1.RT-qPCR and Western blot analysis showed that the expression levels of CEA,NSE and CYFRA21-1 in different lung cancer cell lines.In A549 cells,the protein expression levels of CEA,NSE and CYFRA21-1 were significantly increased after treatment with E2 compared with the blank control group(p=0.023,0.005,and0.001)and the protein expression of CEA,NSE and CYFRA21-1 was significantly decreased after treatment with Ful(p=0.001,0.002,0.001).Similar results were found in H1975,H460 and SK-MES-1 cells.Conclusion This study demonstrates a correlation between the expression of E2,CEA,NSE,CYFRA21-1 and the clinical pathology of patients.The combined detection of E2 and tumor markers is of great significance in the diagnosis of lung cancer.E2 is positively correlated with the expression of tumor markers in lung cancer cells,and patients with high expression of E2 and tumor markers have a poor prognosis.The combined detection of E2 and tumor markers has a guiding role in the prognosis of lung cancer patients.this study provides a basis for the interaction of E2 and tumor markers in lung cancer both in human specimens and cell ? experimental aspects,and its specific molecular mechanism needs further study.
