| Objective:Among the malignant tumors that seriously endanger women’s health and life,breast cancer is the most common seen in disease and has attracted widespread attention from scholars around the world.In recent years,the incidence of breast cancer in China has continued to rise,and the age of onset has gradually become younger.According to the research data,there are about 200,000 new cases of breast cancer every year in China,and about 1.6 million new cases of breast cancer occur every year in the world.Among them,about half a million deaths due to breast cancer deaths every year.In recent decades,with the development of science and technology,the operation effect of breast cancer has made great progress.However,nearly 30% of patients still have recurrence and metastasis every year after surgery.The most important factors are the recurrence and metastasis of the tumor leading to death after breast cancer surgery.Therefore,the study of indicators related to breast cancer recurrence and metastasis is significant for assessing the prognosis of patients.With the emergence of new technologies,the understanding of breast cancer has gradually refined,and the search for tumor markers that help early diagnosis,targeted therapy,and assessment of prognosis has gradually become a hot topic in current research.Most studies on the relation between p120 and CD34 and breast cancer at home and abroad have detected p120 and CD34 in specimens by immunohistochemistry.CD34 is currently recognized as a sensitive vascular marker.By immunohistochemistry,tumor microvessel density(MVD)can be detected.Therefore,MVD value can represent the expression of CD34,which quantitatively reflects tumor microvascular proliferation.The result is consistent,suggesting that p120 and CD34 were positively expressed in breast cancer tissues.The clinical stage,histological grade,and lymph node metastasis of the tumor are closely related,which proves that p120 and CD34 are important in the pathogenesis of breast cancer.However,in recent years,there are few studies on the correlation between p120 and CD34 in breast cancer at home and abroad.A more solid theoretical basis is needed to confirm the relationship between the two.In this study,we examined the expression level of p120 and CD34 in breast and breast fibroadenomas,and explored the relationship between the expression level of p120 and CD34 with clinical and pathological factors,and the correlation between the two.Judging from the role of p120 and CD34 in the pathogenesis of breast cancer,provide the necessary theoretical basis for the initial diagnosis,treatment and prognosis evaluation of breast cancer patients.Methods:1 Clinical data1.1 Experimental group: From September 2017 to June 2018,97 patients with modified breast cancer were enrolled in Four Surgery Department of our hospital,aged 24 to 88 years,with a median age of 50.0 years.The inclusion criterias were as follows: 1 The patients enrolled were female.2 Pathology confirmed as breast cancer.3 No preoperative radiotherapy,chemotherapy,endocrine therapy and targeted therapy.4 medical records and clinical and pathological data are true and complete.5 Obtain patient consent and sign informed consent.6 The experiment was approved by the ethics committee of our hospital.Exclusion criteria: 1 history of other systemic tumor diseases besides breast cancer.2 double breast cancer.1.2 Control group: 78 patients with breast fibroadenoma disease at the same time.2 Method2.1 Detection methods of p120 and CD34: Immunohistochemical method was used to detect the expression of p120 and CD34 in the experimental group and the control group,and the difference of p120 and CD34 expression levels between the experimental group and the control group was compared.2.2 Relationship between p120 and CD34 levels and clinical factors: The relationship between the expression of p120 and CD34 in the experimental group and tumor size,histological grade,lymph node metastasis and TNM staging were analyzed.The following clinical data were collected for each patient enrolled.(1)Tumor size: According to the largest diameter of the tumor,it was divided into <2cm and ≥2cm.(2)Histological classification: Grade I,Grade II and Grade III.(3)Lymph node metastasis: no lymph node metastasis,lymph node metastasis.(4)TNM staging: Phase I,Phase II and Phase III.The above data were collected from clinical medical records and pathological reports.Tumor size,histological grade,and lymph node metastasis were based on postoperative pathological findings.2.3 Correlation between expression of p120 and CD34 in breast cancer tissues: Compare the expression of p120 and CD34 in the experimental group and explore the correlation between the two.3 Statistical methods: The relevant data of this subject is processed by SPSS21.0 software.The measurement data were expressed as mean standard deviation(x ±S).After the normality test,the data between groups that did not conform to the normal distribution were compared by Mann-Whitney U test.The chi-square test was used to compare the count data.The test level was P<0.05,which was statistically significant.The correlation between the two indicators was treated by Spearman rank correlation analysis,where α=0.05,P<0.05 was considered to be statistically significant.Results:1 Expression of p120 and CD34 in breast cancer and breast fibroadenoma: 78 cases(80.4%)were positive for p120 in 97 breast cancer patients,and 10 cases were positive for p120 in 78 patients with breast fibroadenomas.(12.8%).The difference between the two groups was statistically significant(X2=79.012,P<0.05).The MVD of breast cancer patients(25.70±14.37)was significantly greater than that of breast fibroadenomas.The difference between the two groups was statistically significant(P<0.05).2 The relationship between the expression of p120 and CD34 in breast cancer and clinicopathological factors: the relationship between the expression of p120 and CD34 and the clinicopathological factors are shown in the attached table.The proportion of p120 positive expression in patients with different clinical pathological factors showed that the expression of p120 in patients with different tumor sizes was not statistically significant(P>0.05);patients with histological grade III appeared p120 The proportion of positive expression was significantly higher than that of histological grades I and II;the proportion of positive expression of p120 in lymph node metastasis was significantly higher than that in patients without lymph node metastasis;the proportion of positive expression of p120 in stage III TNM patients Significantly higher than TNM I,II,the difference was statistically significant(P<0.05)。The MVD(29.46±12.27)in patients with tumor ≥ 2cm was significantly higher than that in patients with tumor<2cm(19.87±15.57),and the MVD(33.93±10.80)in patients with histological grade III was significantly higher than that in histological patients with grade I and II(18.58±13.29),MVD(32.33±11.79)was significantly higher in patients with lymph node metastasis than in patients without lymph node metastasis(16.25±12.37),and MVD(35.13±11.09)in stage TNM stage was significantly higher than TNM I and II stage MVD(16.47±10.81),the difference was statistically significant(P<0.05).3 Correlation of p120 and CD34 expression in breast cancer: 78 cases of positive expression of p120 in 97 breast cancer patients,of which 45 cases of CD34 expression were also positive,in 19 patients with normal expression of p120 Two cases of CD34 were positive,and the Spearman nonparametric correlation test showed:(r=0.375,p<0.05),indicating that the expression of p120 and CD34 was positively correlated in breast cancer tissues.Conclusion:1 The expression levels of p120 and CD34 in breast cancer patients were significantly higher than those in breast fibroadenomas.It can be concluded that p120 and CD34 are closely related to the occurrence and development of breast cancer.2 Both p120 and CD34 were positively expressed in the breast cancer tissues,and were closely related to tumor differentiation,lymph node metastasis and TNM staging of breast cancer,and could be used as biological indicators for breast cancer metastasis and invasion.3 There is a significant positive correlation between the expression of p120 and CD34 in breast cancer tissues,and it is closely related to the invasion and metastasis of breast cancer.Combined detection is of great significance in evaluating the prognosis and clinical treatment of patients,and it is expected to become a new target for breast cancer treatment. |
