Objective:Breast cancer is one of the highest incidence rate malignant tumors in feminine disease,approximately composes the whole body malignant tumor 7%~10%,its occurrence and development is closely related to protein expression or deletion.The aim of this study is to observe the expression of Syk (Spleen tyrosine kinase) and Survivin protein which is in the family of inhibitor of apoptosis in mammary benign tumor, mammary cancer and normal tissue by immunohistochemical method, the relationship between the expression of Syk and Survivin is analyzed with pathological characteristics,such as pathological stages,tumour size ,lymph node metastasis and etc in breast cancer , the correlation trend is investigated between the expression of Syk and Survivin, these provide evidence for breast cancer of early diagnosis and prognostic evaluation.Methords : 1 70 paraffin blocks of surgery female mammary gland were randomly selected from the first hospital of Qinhuangdao from June of 2006 to June of 2007,including 50 mammary infitrating ductal carcinoma,10 fibroadenoma of mammary and 10 mammary normal tissues.Clinical and pathological data of all patients were collected. The expression of Syk and Survivin were detected by immunohistochemical method , the results were evaluated by the method of semiquantitative scores as Frank[1].The difference between expression of Syk and Survivin was compared in mammary benign tumor,mammary cancer and normal tissues, the relationship of Syk and Survivin was analyzed with pathological characteristics of tumor, the trend of Syk and Survivin was also analyzed.2 Statistical method:Use the SPSS14.0 software system to analyze the results,applicate the rank sum test,linear trend,χ2 test and Spearman grade-related test , analyze the relationship between the expression of Syk and Survivin and pathological characteristics.Results:1 The positive rate of the expression of Syk in breast cancer (78.00%) is lower than that in fibroadenoma of mammary (100%)and normal tissues(100%),the difference has statistical significance(P=0.01).There is no statistical significance on positive rate of the expression of Syk between fibroadenoma of mammary and normal tissue (P>0.05); the positive rate of the expression of Survivin in breast cancer(94.00%) is higher than that in fibroadenoma of mammary (40.00%) and normal tissue (40.00%),the difference has statistical significance(P=0.03).There is no statistical significance on positive rate of the expression of Survivin between fibroadenoma of mammary and normal tissue (P> 0.05).2 The positive rate of the expression of Syk in the no lymph node matastasis group (96.40%)is higher than that in the lymph node matastasis group (55.00%)in breast cancer,the expression of Syk has significant inverse correlation with the lymph node matastasis ( P=0.00);the strong positive rate of the expression of Survivin increases along with the lymph node matastasis numbers gradually,but there is no statistical significance between the expression of Survivin and the lymph node matastasis(P>0.05).3 The positive rate of the expression of Syk declines with the tumor diameter increase gradually,the positive rate of the expression of Syk in group of diameter>5cm (33.30%) is lower than that in the group of diameter≤2cm (85.70%), but there is no statistical significance between them (P>0.05);the positive rate and the strong positive rate of the expression of Survivin increases gradually with the tumor diameter improving,the positive rate of the expression of Survivin in the group of diameter≤2cm (90.50%) is lower than that in the group of diameter>5cm (100%), but there is no statistical significance between them (P>0.05).4 The positive rate of the expression of Syk increases with the positive rate of the expression of TopoⅡ,the positive rate of the expression of Syk in TopoⅡ"-"group(60.00%) is lower than that in the TopoⅡ"++"group(100%),but there is no statistical significance between them (P>0.05);the positive rate and the strong positive rate of the expression of Survivin increases with the positive rate of the expression of TopoⅡ,the positive rate of the expression of Survivin in TopoⅡ"-"group (90.00%) is lower than that in the TopoⅡ"++"group(100%),but there is no statistical significance between them (P>0.05) .5 The positive rate and the strong positive rate of the expression of Syk in the pathological stages ofⅠ-Ⅱgroup(94.10%,64.71%) is higher than that in the pathological stagesⅢ-Ⅳgroup (43.80%,18.75%)obviously(P=0.00); the positive rate and the strong positive rate of the expression of Survivin in the pathology stages ofⅠ-Ⅱgroup(91.20%,76.47%)is lower than that in the pathology stages ofⅢ-Ⅳgroup(100%,100%),(P=0.04).6 There is no statistical significance in the expression of Syk and Survivin in the menopause group and the non-menopause group (P>0.05).7 The positive rate of the expression of Sky increases gradualy along with the increasing of the positive rate of the expression of Survivin, but there is no statistical significance between them (P>0.05).Conclusions:1 The expression of Sky and Survivin is differenced in breast cancer, fibroadenoma and normal tissue.It is a low expression of Sky in breast cancer, and a high expression of Survivin in breast cancer.2 The expression of Syk has significant inverse correlation and linear trend with the lymph node matastasis;the expression of Syk in pathological stage ofⅠ-Ⅱgroup is higher than that in theⅢ-Ⅳgroup;there is no correlation between the expression of Syk and tumor diameter ,the expression of TopoⅡ, menstruation condition.3 The expression of Survivin in the pathological stages ofⅠ-Ⅱgroup is lower than that in theⅢ-Ⅳgroup in breast cancer;there is no correlation between the expression of Survivin and tumor diameter ,lymph node matastasis, the expression of TopoⅡ, menstruation condition.4 There is no correlation between the expression of Syk and Survivin in breast cancer.
