Introduction:Lung cancer is the most serious malignant tumor threatening human health and life,and is an important cause of cancer-related deaths worldwide.The five-year survival rate is only 15%,and the incidence rate is gradually increasing.Most patients with lung cancer have no obvious clinical manifestations and signs in the early stage,and the prognosis is relatively poor.Recent studies have confirmed that immune escape is one of the important mechanisms for the occurrence and development of malignant tumors.The occurrence,development,metastasis and prognosis of lung cancer are also the result of tumor cells escaping from the body's immunity and even attacking the immune system.The most important is the effect of CD4+T cells and related cytokines on their function.Different cytokines induce the production of different transcription factors and stimulate the expression and differentiation of initial CD4+T cells into regulatory T cells(Treg)and helper T cells 17(Th17).Their functions and phenotypes are different.CD4+T cell subsets are key participants in various diseases involving inflammation,autoimmune diseases and tumors.Th17 and Treg mainly regulate the immune response of cells.The differentiation and development regulation mechanism of Th17 mainly secretes il-17 specifically,which is involved in the occurrence and development of various diseases in the body and is closely related to their prognosis and prognosis,including inflammatory diseases,autoimmune diseases,malignant tumors and immunosuppression.Treg mainly plays a role in inhibiting epidemic diseases.The surface marker Foxp3,which non-specifically inhibits the activation of Th17,reduces its expression and thus inhibits its function.Treg surface there are other more important surface marks,including CD25 molecule and cytotoxic T lymphocyte antigen 4(CTLA-4),and CD8,CD4+ CD25 + Treg in normal human peripheral blood CD4+ T cells in 5%-10%,dynamically adjust the body's immune balance,make the body's immune environment stable,when the number of Treg reduce or insufficient function,the body's immune steady state may be destroyed,there will be a slight immune damage,serious when even in autoimmune diseases,populations,or enhancements,the immune function of organism is restrained,This is one of the reasons for the formation of tumor immune tolerance,which weakens the immune response of the body's immune system to tumor cells and promotes the occurrence and development of tumor.These two types of CD4+T cells have a subtle relationship with the occurrence,progression and metastasis of tumors,which is of important research value.Method:Flow CytoMetry(this study applies the Flow CytoMetry,FCM)detection in the peripheral blood of Th17 and Treg in NSCLC patients compared with healthy subjects peripheral blood levels of two kinds of cells in the comparison of NSCLC patients cured before 1 day,1 day after operation,1 week after the change trend of these two kinds of cells in peripheral blood,compare different pathological stages of peripheral blood,the content of two kinds of cells in NSCLC patients to explore these two kinds of cells with NSCLC occurrence,development,metastasis and recurrence,the relationship between the change of dynamic monitoring of the peripheral blood cells content trend to evaluate surgical curative effect,It provides a basis for further postoperative treatment and assessment of prognosis,and provides a theoretical basis for immunotherapy of NSCLC and even other malignant tumors.Results:Th17:1.The content of Th17 in peripheral blood of NSCLC patients was higher than that of healthy people(P<0.05).2.There was no significant difference in the content of Th17 in peripheral blood of NSCLC patients of different genders and ages(P>0.05).3.The higher the TNM stage,the lower the content of Th17 cells in the peripheral blood of NSCLC patients(P<0.05).4.The expression of Th17 in peripheral blood was significantly reduced 1 day after radical resection of NSCLC compared with that on the 1 day before surgery(P<0.05),and significantly increased 1 week after surgery compared with that on the 1 day after surgery(P<0.05).Treg:1.The expression of Treg in peripheral blood of NSCLC patients was higher than that of healthy subjects(P<0.05).2.There was no significant difference in the content of Treg in peripheral blood of NSCLC patients of different genders and ages(P>0.05).3.The higher the TNM stage,the higher the content of Treg cells in peripheral blood(P<0.05).4.Treg in peripheral blood decreased significantly 1 day after radical resection of NSCLC compared with 1 day before surgery(P<0.05),and increased significantly 1 week after surgery compared with 1 day after surgery(P<0.05).Treg/Th17:1.There was no difference in the proportion of Treg/Th17 in peripheral blood of NSCLC patients of different genders and ages(P>0.05).2.The higher the TNM stage,the higher the proportion of Treg/Th17 in peripheral blood(P<0.05).3.There was no statistically significant difference in Treg/Th17 before and after radical resection of NSCLC(P>0.05).Conclusion:1.High levels of Th17 and Treg in peripheral blood are correlated with the occurrence of NSCLC.2.The contents of Th17 and Treg in peripheral blood of NSCLC patients were not correlated with age and gender,but correlated with tumor TNM staging.3.Radical resection of NSCLC can reduce the contents of Treg and Th17 in peripheral blood of patients.4.The dynamic monitoring of the expression of Th17 and Treg in peripheral blood is of certain significance for the formulation of the follow-up immunomolecular targeted therapy.
