Objective:Bacterial Vaginosis?BV?is a common disease in fertile women and is associated with potentially serious side effects,including an increased risk of preterm birth.With the development of microbial sequencing technology,new research has been made on the complex mechanism of BV,and some new diagnostic methods have been derived.In this study,12K Flex Real-Time PCR technology and Next Generation Sequencing?NGS?was used to detect and analyze vaginal flora in 17 patients with intermediate BV,26 patients with bacterial vaginosis and 19 healthy subjects.The main purposes of this study are:first,to identify the pathogens in the vagina of patients with BV;second,to analyze the distribution of vaginal flora in intermediate BV,BV patients and healthy people to obtain a more comprehensive and accurate understanding of the distribution of vaginal flora.It provides a scientific basis for the discussion of the etiology of BV and the prevention and treatment of mixed infection.Methods:From August 2017 to December 2017,the samples from BV patients and normal persons in the gynaecological specialist clinic of the First Affiliated Hospital of Xi'an Medical College were selected as the experimental subjects.The vaginal secretions of the patients were taken as experimental samples,and the vaginal microecological detection method was used to diagnose BV.?1?Species identification of microorganisms commonly found in vaginal flora using QuantStudioTM 12K Flex Real-Time PCR detection technology;?2?Detection of bacterial diversity of vaginal flora based on 16S rDNA amplicon sequencing of V3-V4 region.Results:?1?The structure of the flora in the vagina of healthy people is relatively simple,the species richness is low,and the bacterial structure of BV patients is complex,while the species richness is much higher than that of normal people.?2?PcoA analysis showed that there was no significant difference in the distribution of bacterial flora between intermediate BV and BV patients.In the genus heat map distribution,the two groups of samples can be clearly distinguished,and the healthy group can be clearly distinguished from the BV group and the suspected BV group.?3?The dominant bacteria in the healthy group are mainly Lactobacillus,while the dominant bacteria in BV patients are mainly Gardnerella,Prevotella,Sneathia,Megasphaera,Atopobium,Streptococcus,Prevotella-7,and Mycoplasma.?4?QuantStudioTM 12K Flex Real-Time PCR detection technology is basically consistent with the dominant bacteria detected by NGS technology,and 12K Flex Real-Time PCR detection technology can identify species.Conclusion:Both QuantStudioTM 12K Flex Real-Time PCR detection technology and NGS technology have their own advantages.However,12K Flex Real-Time PCR detection technology can specifically identify the level of the species.The vaginal flora of BV patients has higher species diversity,the composition of the flora is complex,and the dominant flora is dominated by anaerobic bacteria.There was a significant difference in vaginal flora between intermediate BV and BV patients.Lactobacilli of intermediate BV are more abundant than BV patients.The vaginal flora of healthy people has low species richness and simple flora,mainly Lactobacillus.
