Objective:To investigate the effect and mechanisms of ?7 nicotine Acetylcholine receptor(?7nAChR)PNU-282987 on IL-4 induced M2 polarization in MH-S.Method:The MH-S murine alveolar macrophage cell line were incubated and sorted into groups as follows:1.MH-S cells were stimulated by PNU-282987(0.1?M,1?M,10?M,100?M,300?M,500?M)or vehicle for 30 min before stimulated with IL-4(10ng/ml).Then the expression of Arg1,Ym1 and CD206 were examined by RT-PCR and Western Blot.2.To confirm the mechanism of PNU-282987 and IL-4 on the expression of Arg1,Ym1 and CD206,MH-S were treated by the antagonist of STAT6(leflunomide)or PPAR gamma(GW9662)before PNU-282987 and IL-4 stimulation.The expression of STAT6,nucleus PPAR gamma,Arg1,Ym1 and CD206 were examined by RT-PCR,Western Blot and Flow Cytometry.Result:PNU-282987 promotes IL-4 induced MH-S M2 polarization at 100?mol/L.The expression of STAT6 and PPAR gamma were significantly improved at 2h after IL-4 treatment.And inhibition of STAT6 and PPAR gamma inverts the enhancement of the expression of STAT6,nucleus PPAR gamma,Arg1,Ym1 and CD206.Conclusion:PNU-282987 promotes IL-4 induced MH-S M2 polarization via activating the expression of STAT6 and PPAR gamma.
