Objectives To explore the ability of restoration and regeneration of to dental pulp of the new bioactive material,hyaluronic acid-silver bioactive glass composite(HA/Ag-BG),in order to obtain an ideal direct capping material and provide experimental basis for clinical application.Methods Twenty-one adult male Sprague dawley(SD)rats,weighing(220±10)g,were housed in a specific pathogen free(SPF)environment,and the rats were fed with sterile water and feed.Twenty-one rats were randomly divided into HA/Ag-BG group,Mineral Trioxide Aggregate(MTA)group and Calcium hydroxide(CH)group,with 7 rats in each group.Four primary molars were selected as the study subjects in each SD rat.In the case of general anesthesia,a high-speed turbine was used to drill the proximal molars of the first molar in the rat to the near pulp,and the microprobe was used to probe.A refoldable pulpitis model was established by applying a solution of lipopolysaccharide(LPS)at a concentration of 5?g/?l after opening the marrow.After the model was established,the cells were directly covered with HA/Ag-BG,MTA,and CH,and cultured for 8 weeks.After 8 weeks,the experimental rats were sacrificed by abdominal anesthesia.The upper and lower jaws were separated,specimens were obtained,histological sections were prepared,and hematoxylin and eosin(HE)staining was performed to observe the intramedullary inflammation and repair dentin.In the case of bridge formation,statistical analysis was performed and statistical analysis was performed using rank sum test.Dentin sialoprotein(DSP)and vascular endothelial growth factor(VEGF)immunohistochemistry were performed.The results were analyzed by Image-Pro Plus 6.0 software.The data were analyzed statistically.Results 1.Inflammatory reaction of SD rats after direct pulp capping: 8/25(32%)specimens in the HA/Ag-BG group had mild inflammation,and 13/25(52%)specimens showed severe inflammation in the pulp.In the reaction,4/25(16%)specimens had abscess necrosis in the pulp;in the MTA group,16/24(66%)specimens showed mild inflammation in the pulp,and 4/24(16%)specimens had severe inflammation in the pulp.4/24(16%)specimens showed disintegration and necrosis in the pulp;in the CH group,4/28(14%)specimens had mild inflammatory reaction in the pulp,and 10/28(36%)specimens showed severe inflammatory reaction in the pulp.Abscess necrosis occurred in14/28(50%)specimens;statistical analysis showed no significant difference between HA/Ag-BG group and MTA group(P>0.05),that is,HA/Ag-BG and MTA had similar anti-inflammatory effects..The difference between HA/Ag-BG group and CH group was statistically significant(P<0.05),that is,the anti-inflammatory effect of HA/Ag-BG and MTA was better than CH.2 The repairing dentin formation of SD rats after direct pulp capping: 3/25(12%)of the HA/Ag-BG group had intact calcified bridge formation at the perforation,10/25(40%)specimens No formation of intact calcification bridge was observed at the perforation,but calcification was observed near the perforation.No calcified calcification was observed in the perforation of 12/25(48%)specimens;14/24 in the MTA group(58%)There was a complete calcification bridge at the perforation of the specimen,and no complete calcification bridge was formed at the perforation of the 6/24(31%)specimen,but calcified mass formation was seen near the perforation,8/24(17%)no calcified tissue was formed at the perforation of the specimen;1/28(4%)of the CH group had a complete calcification bridge at the perforation,and 5/28(18%)specimenswere near the perforation.When calcified mass formation was observed,the remaining22/28(78%)specimens were free of calcified tissue at the perforation;statistical analysis showed that there was a statistically significant difference between the HA/Ag-BG group and the MTA group(P<0.05).The ability of HA/Ag-BG to form a repairing dentin bridge was lower than that of the MTA group;there was no significant difference between the HA/Ag-BG group and the CH group(P>0.05),that is,HA/Ag-BG formed a repaired tooth.The ability of the nature bridge is similar to CH;3 HA/Ag-BG expression of DSP and VEGF in the pulp after direct pulp capping: DSP and VEGF in the medullary cavity of MTA group showed strong positive expression,HA/Ag-BG and CH group showed positive expression,HA There was significant difference in DSP expression between the /Ag-BG group and the MTA group(P<0.05),that is,the DSP expression in the MTA group was stronger than that in the HA/Ag-BG group.There was no significant difference in DSP expression between HA/Ag-BG group and CH group(P>0.05).The HA/Ag-BG group,MTA group and CH group were significantly different from the blank control group(P<0.05),that is,HA/Ag-BG,MTA and CH could promote DSP expression;HA/Ag There was no significant difference in the expression of VEGF between the BG group and the MTA group and the CH group(P>0.05),but the differences between the HA/Ag-BG group,the MTA group and the CH group were statistically significant.(P<0.05),that is,HA/AgBG,MTA,and CH all promoted the expression of VEGF.Conclusions 1 HA/Ag-BG has good inflammatory control effect,which is similar to that of MTA,and stronger than CH;2 The ability of HA/Ag-BG to induce dentin regeneration is lower than that of MTA
