Objective To understand the pathogenesis of wind-heat sores by studying the correlation between human leukocyte antigens and wind-heat sores.It is of great significance to prevent the occurrence of this disease and reduce the incidence rate.It also provides the basis of immunology for clinical treatment and provides a scientific basis for achieving precision medicine.On the other hand,studying the correlation between TCM syndrome type of wind-heat sores and HLA helps us to understand the essence of dialectical classification of the certificate of Chinese medicine from the genetic level,and to provide a more accurate view of TCM syndrome type differentiation of wind-heat sore from a microscopic point of view.Methods By collecting and sorting out the relevant literature,63 cases of wind-heat sore were selected according to the diagnostic criteria of Chinese and Western medicine symptoms.dialectical classification of Chinese medicine dialectics is divided into two types: Wind-heat skin accumulation type and Wind-heat and blood-dryness type.32 cases of Wind-heat skin accumulation type patients,31 cases of wind-heat and blood dryness,and another 100 healthy persons were selected as the normal control group.The case group and the control group are both Inner Mongolian nationals.The blood samples were collected,DNA was extracted,and HLA-DQB1 and HLA-DRB1 genes were classified by multiple PCR technology and Illumina second-generation sequencing technology.The results of typing were analyzed by chi-square test.The correlation between HLA-DQB1 and HLA-DRB1 alleles in patients with rheumatic fever and healthy persons was discussed.Whether the allele is relevant between the case group and the control group and between the syndrome types is also under discussion.Results 1.It was found that the frequency of HLA-DRB1*11:04 in patients withrheumatic fever was higher than that of the normal control group(3.97%vs0.5%,P=0.033,P < 0.05;OR=8.176,95%CI: 0.899-390.058),the frequency of its high-resolution six-digit HLA-DRB1*11:04:01 in patients with wind-heat sores is also higher than the normal control group(5%vs1%,P=0.043,P<0.05;OR=7.453,95%CI: 0.819-356.077)and the frequency of HLA-DRB1*07:04 in patients with rheumatic fever was lower than that of the normal control group(0.79%vs5.5%,P=0.032,P<0.05;OR=0.138,95%CI: 0.003-0.971).2.For the wind-heat sore,the gene frequency of HLA-DRB1*11:04 is higher than that of the healthy control group(7.81%vs0.5%,P=0.003,P < 0.05;OR=16.646,95%CI: 1.812-797.884).By comparing wind-heat skin accumulation type and wind-heat blood dryness type of wind-heat sore in dialectical classification of Chinese medicine,there was no difference between HLA-DRB1*11:04 and HLA-DRB1*07:04 alleles between the two types of syndromes.Conclusions 1.The results showed that HLA-DRB1*11:04 may be a susceptibility gene in the pathogenesis of wind-heat sores,and HLA-DRB1*07:04 may be a protective gene.2.Compared with the healthy control group,HLA-DRB1*11:04 may be a susceptibility gene of wind-heat accumulation syndrome in wind-heat ulcer.
