Objective: The aim of this study was to observe the effect of Gambogic acid derivative compound 5 on apoptosis of human oral squamous cell carcinoma(OSCC)in vitro.Materials and methods: In the present study,oral squamous cell carcinoma cell lines CAL27,HN13 and HN30 were treated with C5 at different doses and different times.The cell viabilities were detected with MTT assay and the IC50 values were also determined by Logit method.Apoptosis induced by C5 was observed by Annexin-V/PI doubling staining in the flow cytometry assay in three cell lines.The change on the expression of apoptosis-related proteins was detected by Western blot and fluorescent immunocytochemistry assay.Results: CAL27,HN13 and HN30 cell lines were dramatically inhibited in a dose-dependent and time-dependent manner after incubation with C5.The IC50 values were much less than the reference drug GA.When CAL27 cells were incubated with high dose C5 of 5.0 ?mol/L for 24 h,48 h and 72 h,the apoptotic population was about 52.19%,65.24% and 84.53%,respectively.C5 could decrease protein level expression of Bcl-2 and increase protein level expression of Bax in CAL27,HN13 and HN30 cell lines.Conclusions: Altogether,our data demonstrated that C5 induced apoptosis of OSCC cell lines with up-regulation of Bax and down-regulation of Bcl-2 in vitro.
