Effects Of Niuhuang Jiangya Capsule On Blood Pressure And Cardiac And Renal Function In Spontaneously Hypertensive Rats

Niuhuang Jiangya formula is a classic antihypertensive formula,with functions such as clearing heart,removing blood stasis and reducing blood pressure.It has a definite curative effect on hypertension.In present,the Niuhuang Jiangya preparations in clinical application include pills,tablets and capsules.There are quantities of clinical reports on Niuhuang Jiangya pills,while the clinical reports on Niuhuang Jiangya capsules are relatively few,and experimental research is even less.The purpose of this study was to evaluate the antihypertensive effect,characteristics and safety of Niuhuang Jiangya capsule on spontaneously hypertensive rat(SHR)model.On the basis of revealing Niuhuang Jiangya capsule potential protective effects on spontaneous hypertensive cardiovascular complications,and investigate its antihypertensive mechanism,to provide experimental basis for clinical application of Niuhuang Jiangya capsule.Part One The effect of Niuhuang Jiangya capsule on blood pressure in spontaneously hypertensive ratsObjective: To evaluate the hypotensive activity and hypotensive characteristics of Niuhuang Jiangya capsule,and to compare the advantages and disadvantages of three different kinds of Niuhuang Jiangya preparations.Methods: Total 32 male spontaneously hypertensive rats were randomly divided into 4 groups,8 rats in each group,and were set as model control group(A group),Niuhuang Jiangya capsule group(B group),Nuhuang Jiangya pill group(C group),and Niuhuang Jiangya tablet group(D group),respectively,while another 8 WKY rats was set as normal control group.The dosage of Niuhuang Jiangya capsule,Niuhuang Jiangya pills and Niuhuang Jiangya tablet group were 0.32 g/kg,0.64g/kg and 0.30g/kg weight,respectively.After continuous administration for 31 days,blood pressure was measured at 2 h every 5 days after the end of administration.Blood pressure of 2 h,6 h and 24 h was measured 10 days later.Blood pressure of 2 h,6 h,12 h,24 h and 48 h was measured after the last administration.Results: Niuhuang Jiangya capsule in the treatment for sixth days showed antihypertensive effect,after 21 days of treatment reached the maximum antihypertensive effect,and antihypertensive effect at different time points within a day keeps stable,administered for 31 days after cessation of dosing,48 h remained a significant antihypertensive effect.Niuhuang Jiangya capsule and other forms of comparative antihypertensive effect there was no statistical difference,but works earlier than Niuhuang Jiangya pill.Conclusion: Niuhuang Jiangya capsule can effectively reduce the blood pressure of spontaneously hypertensive rats with the characteristics of stable and long-lasting effect.Part Two The effect of Niuhuang Jiangya capsule on cardiac and renal function in spontaneously hypertensive rats.Objective:(1)To evaluate the damage of liver and kidney function by long term treatment with biochemical markers of liver and kidney function combined with pathological results.(2)To evaluate the effect of hypertension model on heart,liver,kidney and spleen by pathological means,and to evaluate the preventive and therapeutic effects of Niuhuang Jiangya capsule on complications such as myocardial hypertrophy and renal arteriolar sclerosis.(3)To determine the levels of renin and angiotensin II in blood and kidney,and preliminarily reveal the mechanism of Niuhuang Jiangya capsule.Methods: After administration of 48 h,the normal control group(WKY group),model group(A group),Niuhuang Jiangya pill group(B group)were killed,and then the animal heart,kidney,spleen and liver were weighed and fixed in 10% formalin.After dehydration,embedding,sectioning and sealing,hematoxylin-eosin(HE)staining was performed for pathological analysis.The levels of renin and angiotensin II in the kidney and plasma of each group were determined by ELISA kit.Results: The pathological results showed that continuous intragastric administration for 31 days,the drug group showed no pathological changes of liver and spleen,and the normal control group and model group had no significant difference.Niuhuang Jiangya capsule on hypertension induced myocardial thickening and glomerular expansion has a preventive effect.Niuhuang Jiangya capsule can significantly reduce the levels of renin and AngII in the plasma of hypertensive animals.Conclusion: There was no obvious adverse reaction in the long-term use of Niuhuang Jiangya capsules.Niuhuang Jiangya capsule has a certain preventive effect on heart and kidney complications caused by hypertension.The hypotensive mechanism of Niuhuang Jiangya capsule may be related to the reduction of renin and AngII.
