Primary biliary cholangitis(PBC)is an autoimmune-mediated cholestatic,chronic inflammatory liver disease,formerly known as primary biliary cirrhosis.PBC mostly occurs in middle-aged women,approximately 50(median)years old,with a male to female ratio of up to 1:9.The common clinical symptoms of the disease are fatigue,itching,etc.Besides,other autoimmune diseases are also usually combined with PBC.Studies have shown that the disease progression of PBC is related to early diagnosis,so early diagnosis of PBC is very important to patients.At present,the biological markers for clinical diagnosis of PBC mainly include anti-mitochondrial antibody-M2(AMA-M2)and anti-nuclear antibody(ANA).In clinical test of PBC,ANA mainly includes anti-Sp100 and Anti-Gp210.Although the positive rate of AMA-M2 can reach 95% in PBC patients,there are still 5% of patients with PBC who are negative for this antibody.At this time,ANA detection is very important for these 5% of PBC patients.In order to further improve the diagnostic rate of AMA-M2-negative PBC patients,it is a good method to add new relevant biological markers.Recent years,a new kind of autoantigen named small ubiquitin-related modifier(SUMO)has been found in PBC.It is reported that anti-SUMO antibody has been confirmed as a new specific antibody in Caucasian PBC,so this study aims to explore whether the anti-SUMO antibody can also become a new specific antibody in Han Chinese PBC.In addition,according to report,anti-SUMO antibody is associated with anti-nuclear dots(anti-NDs)in Caucasian PBC,and anti-Sp100 antibody is an important one of anti-NDs.Therefore,my study would also explore the relationship between anti-SUMO antibody and anti-Sp100 antibody in Han Chinese PBC and determine whether anti-SUMO antibody can be an accessory molecule for diagnosis of AMA-M2-negative PBC.In this study,three SUMO proteins(SUMO1/2/3)are prokaryotically expressed and purified by molecular biology methods.Some PBC cases who show anti-SUMO antibody positive are obtained by dot blot and Western blot,and the anti-SUMO antibody ELISA test method is initially established to test the serum samples of 319 PBC cases and 416 other autoimmune disease(including systemic lupus erythematosus,rheumatoid arthritis,and Sjogren's syndrome)patients.Finally,relevant statistical analysis is performed.Statistical analysis shows that there is a statistically significant difference in the positive rate of anti-SUMO antibody(anti-SUMO1/2/3)in PBC and other autoimmune diseases samples(P value < 0.05),suggesting that anti-SUMO antibody may be a new specific antibody of Han Chinese PBC.Besides,in the anti-Sp100 group of PBC patients,the positive rate of anti-SUMO antibody in the anti-Sp100 positive group is significantly higher than that in the anti-Sp100 negative group(P value < 0.01).Therefore,it may reveal that there is also a certain correlation between anti-SUMO antibody and anti-Sp100 antibody in Han Chinese PBC.
