Stellera chamaejasme L.,a medicinal plant of the traditional Chinese medicine,which is widely distributed in northwest,southwest,northeast and Hebei of China.The traditional Chinese medicine records that it has the effect of treating various diseases.At present,it is found that it has potent anti-tumor and HIV activities.In this study,the chemical constituents of Stellera chamaejasme L.were extracted and isolated.By means various modern chromatographic techniques?normal phase and reverse phase silica gel column,Sephadex LH-20,MPLC,HPLC,etc.?,and modern spectroscopic analysis methods(1H-MNR,13C-NMR,DEPT,HSQC,1H-1H COSY,HMBC,NOESY,MS,etc.),14 compounds were isolated and identified.Their bioassay experiments also were carried out.The main results are shown as follows:14 compounds were isolated from the ethyl acetate extract fraction of Stellera chamaejasme L.:one diterpene orthoester:Stelleralide C?1?,three biflavonoids:chamaechromone?2?,neochamaejasmin B?3?,isoneochamaejasmin A?4?,three flavonoids:epiafzelechin?5?,kaempferol?6?,Naringenin?7?,six lignans:?+?-isolariciresinol?8?,?-?-lariciresinol?9?,syringaresinol?10?,?+?-Matairesinol?11?,?+?-pinoresinol?12?,acutissimalignan B?13?,one coumarin:scopoletin?14?.The biological activities of some of the compounds were tested,including cytotoxic,anti-inflammatory,neurotrophic,and?-glucosidase inhibitory.The results showed that the compound 1,2,3,4,6 showed strong cytotoxic activity against U87MG with IC500 values of 2.8,45.1,25.4,25.6,24.1?M;compounds 1,10 had strong anti-inflammatory activity using BV-2 cell model,the inhibition rates at 1?M concentration were 40.476%,40.596%;all the tested compounds had no neurotrophic activities;and compounds 2,3,4,and 6 all showed strong?-glucosidase inhibitory activity with IC500 values of 24.1,2.99,3.26,and 14.2?M,respectively.
